摘要
目的观察富血小板血浆(PRP)促进纤维多孔钛椎体间融合器(fiber porous titanium Cage,FPT Cage)的骨长入作用。方法健康成年比格犬6只,体质量12.5-15.0 kg,雌雄各3只。取比格犬前肢外周静脉血20.0 m L,采用二次离心法制备PRP,同时对外周静脉血及PRP行血小板计数。行颈椎前路C4/C5/C6椎间盘切除FPT Cage椎间融合术,C4/C5节段单纯使用FPT Cage,C5/C6节段采用激活的PRP浸泡的FPT Cage。术后4个月处死比格犬后行硬组织切片检查,观察C4/C5/C6节段FPT Cage中骨长入的情况。结果 C4/C5节段椎间隙FPT Cage-骨界面附近可见少量软骨细胞和成骨细胞;FPT Cage中央孔隙内发现少量成骨细胞,类骨质较少,大部分孔隙内仍然为纤维组织。C5/C6节段椎间隙FPT Cage-骨界面可见成熟骨组织形成;FPT Cage边缘孔隙内可见骨组织长入;FPT Cage中央孔隙内发现大量成骨细胞;大部分中央孔隙内为类骨组织,少部分中央孔隙内仍然为纤维组织。结论在比格犬体内实验研究中,复合PRP可以促进FPT Cage内的骨长入。
Objective To observe the promotion effect of platelet-rich plasma(PRP) of bone ingrowth in fiber porous titanium Cage(FPT Cage). Methods Six beagle dogs weighing 12.5- 15.0 kg were chosen,3 males and 3 females. Before the discectomy and fusion,20.0 m L fresh blood was obtained from the foreleg of each dog,and PRP was obtained by twostep centrifugation method. Platelet count was performed in peripheral venous blood and PRP. Anterior cervical C4/C5/C6 intervertebral disc resection and FPT Cage interbody fusion were carried out for each dog. FPT Cage was used alone in C4/C5 segment. For C5/C6 segment,FPT Cage augmented with activated PRP were used. The fusion sites were evaluated with scleroustissue-slice technique,and observed under light microscope after staining with toluidine blue 4 months after the operation. Results At the C4/C5 segment,a small number of chondrocytes and osteoblasts were found on the bone-FPT Cage interface;In the central pores of FPT Cage,a small number of osteoblast cells and osteoid can be found;Most of the central pores were filled with fibrous connective tissue. At the C5/C6 segment,mature bone tissue was found on the bone-FPT Cage interface;Bone tissue was also found at the marginal pores of the FPT Cage;A large number of osteoblast cells were found in the central pores of the FPT cage;Most of the central pores were of bone-like tissue,and a small part of the central pores were still fibrous tissue.Conclusion In in-vivo experimental study of beagle dogs,the compound PRP can promote the bone ingrowth in FPT Cage.
