摘要
目的:通过腹腔注射苯妥英钠溶液的方法诱导建立大鼠牙龈增生模型,测量牙龈体积,观察病理学表现,确定诱导大鼠牙龈增生的最佳给药浓度及可靠性,为进一步研究牙龈上皮增生提供实验基础。方法:将60只大鼠随机平均分为对照组(腹腔注射生理盐水1 m L)和实验一、二、三、四组(腹腔注射0、5、25、50、100 mg/m L PHT溶液),连续28d,肉眼观察大鼠牙龈形态改变,取下颌骨标本,测量下前牙牙龈体积,观察牙龈病理表现。结果:100 mg/m L组表现出牙龈增生的大鼠数量最多,牙龈体积变化最为显著。大鼠下前牙牙龈体积组间比较差异显著,且与给药剂量正相关。结论:腹腔注射PHT溶液28d后多数大鼠出现牙龈增生,100 mg/kg组的发病率最高,增生最显著。牙龈增生主要发生于前牙区和下颌第一磨牙区,其组织病理表现与以往研究基本一致。
Objective: To establish rat models of gingival hyperplasia induced by PHT,ensure the best optimum concentration of PHT and reliability through measuring the volume change of gums and observing the pathological changes,in order to provide the experimental basis for the study of gingival epithelial hyperplasia. Methods: 60 SD rats were randomly divided into five groups equally: 0( control group),5 mg / m L,25 mg / m L,50 mg / m L,100 mg / m L injected with PHT. All rats were operated through intraperitoneal injection for 28 days,observed rat morphological change by macroscopic,then sacrificed all the rats for measuring gum volume of under former and observing the pathology of gums through mandibular specimens. Results: 100 mg / m L group showed the largest number of gingival hyperplasia in rats,and the change of gum volume was the most significant. The gum volume of mandibular anterior teeth had significant difference between each group,and positively related to the drug delivery dosage. Conclusion: After intraperitoneal injection of PHT,most gingival hyperplasia in rats appeared in 28 days,the highest rate of gingival hyperplasia happened in the 100 mg / kg group. Gingival hyperplasia mainly happened in the mandibular anterior teeth and mandibular first molar area,its histopathological performance was consistent with previous research.
