活性炭-季铵化壳聚糖澄清剂的制备工艺优化 被引量:3

摘要 制备了以活性炭为载体的季铵化壳聚糖澄清剂,用于中药槐米水提液的澄清处理,以提取液的透光率和黄酮的保留率为响应值,采用响应面法实验设计,考察了活性炭/季铵化壳聚糖质量比、季铵化壳聚糖浓度和超声时间对澄清剂的性能影响。结果表明,制备澄清剂最佳工艺参数为:活性炭/季铵化壳聚糖质量比8∶1,季铵化壳聚糖浓度2%和超声时间20 min。在此条件下,制备的澄清剂处理槐米提取液后的透光率为91.62%,澄清效果较好,黄酮保留率为75.62%,有效成分得到较好的保留。该澄清剂制备工艺简单、成本低廉、绿色环保,可为中药精制工业化应用提供参考。 To prepare the clarifying agent of quaternized chitosan supported by activated carbon to be used for clarification in water extract from Huaimi,the effects of mass ratio of activated carbon to quaternized chitosan,concentration of chitosan and treatment time by ultrasonic were investigated using response surface methodology with the retaining rate of flavonoids and the light transmittance of extracting solution as indexes. The results showed that the optimum conditions for preparation were ratio of activated carbon to quaternized chitosan 8∶ 1,concentration of chitosan 2% and treatment time by ultrasonic 20 min. Under these conditions,the light transmittance was 91. 62%,which indicated that the clarifying agent had a good clarification effect on the extracting solution from Huaimi; and the retaining rate of flavonoids was75. 62%,which showed that the effective component of flavonoids was well preserved during the clarifying process. Owing to its simplicity,cheapness,pollution-free,reliability,the preparation technology of clarifying agent will provide a reference for industrialization utilization of purification in traditional Chinese medicine.
