四妙散及其加减方对高尿酸合并高脂血症大鼠的影响及机理探讨 被引量:5

Effect and Mechanism of Simiao San and Its Modified Formula on Serum Metabolic Substances in Rats with Hyperuricemia Complicated with Hyperlipaemia

摘要 目的研究四妙散及其加减方对高尿酸合并高脂血症大鼠血清尿酸、血脂指标以及黄嘌呤氧化酶(XOD)、黄嘌呤脱氢酶(XDH)、超氧化物岐化酶(SOD)、丙二醛(MDA)的影响。方法 SD大鼠40只,随机分为正常对照组、模型组、四妙散组(10g·kg^(-1),按生药量计)、四妙散加减方组(13g·kg^(-1),按生药量计),每组10只。采用腺嘌呤+氧嗪酸钾+高糖高脂饲料制备高尿酸合并高脂血症大鼠模型。给药30d,戊巴比妥钠麻醉,采血并分离血清测定尿酸(UA)、尿素氮(BUN)、肌酐(Cr)、总胆固醇(CHO)、甘油三酯(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)的水平;取肝组织匀浆测定XOD、XDH、SOD活性及MDA含量,取肾组织匀浆测定SOD活性及MDA含量;取肾脏切片,HE染色观察肾组织形态。结果与模型组比较,四妙散及其加减方组大鼠血清代谢指标均有不同程度的改善;四妙散加减方能显著降低血清UA、CHO、TG水平(P<0.05),升高HDL水平(P<0.05),显著降低肝组织XOD、XDH水平(P<0.05),显著升高肝、肾组织中SOD水平(P<0.05),降低肝、肾组织中MDA含量(P<0.05)。结论四妙散加减方具有降尿酸和降血脂的功效,其机制可能与其抑制XOD、XDH活性及增强SOD活性相关。 Objective To investigate the effect of Simiao San(SS) and its modified formula on serum uric acid(UA),blood lipid, xanthine oxydase(XOD), xanthine dehydrogenase(XDH), superoxide dismutase(SOD) and malondialdehyde(MDA) in rats with hyperuricemia complicated with hyperlipaemia. Methods Forty Sprague- Dawley(SD)rats were randomly divided into 4 groups,normal group,model group,SS group(10 g·kg^(-1)),and modified SS group(13 g·kg^(-1)), 10 rats in each group. Rat model with hyperuricemia and hyperlipaemia was established by the combined use of adenine, oteracil potassium, and high fat and high sugar diet. After medication for 30 days, rats were anesthetized with intraperitoneal injection of pentobarbital sodium,and blood was sampled from abdominal artery for the detection of UA, blood urea nitrogen, creatinine, total cholesterol(TC), triglyceride(TG), high- density lipoprotein(HDL)and low- density lipoprotein(LDL). The activities of XOD,XDH and SOD as well as MDA content in liver tissue homogenate were examined. Histopathological changes in the kidney were observed by HE staining,and SOD activity and MDA content in kidney tissue homogenate were also detected. Results Compared with the model group,serum metabolic parameters in SS group and modified SS group were improved in varied degrees. Modified SS could decrease UA,TC and TG levels,increase HDL,decrease hepatic XOD and XDH,increase hepatic and renal SOD activity, and decrease hepatic and renal MDA content(P〈0.05). Conclusion Modified SS has the action of lowering uric acid and blood lipid, and the possible mechanism is related with the inhibition of XOD and XDH activities and with the enhancement of SOD activity.
