摘要
目的观察百合知母汤提取物(Lily-Zhimu extracted substances,L-ZES)对高脂饲料喂养的抑郁模型大鼠的疗效及作用机制。方法 50只雄性SD大鼠随机分为5组,即K组,正常组;A组,抑郁模型组;B组,抑郁模型百合知母汤给药组;C组,高脂饲养抑郁模型组;D组,高脂饲养抑郁模型百合知母汤给药组。抑郁模型采用慢性不可预见性温和刺激(CUMS)配合孤养诱发。连续模型复制20周,同时灌胃给药,观察大鼠体质量变化,运用旷场实验(the Open Field Tes)t、强迫游泳实验(the forced swim test,FST)评定大鼠的行为学特征。以酶联免疫吸附法(ELISA)检测血浆谷氨酸(GLU)、五羟色胺(5-HT)的含量。结果与正常组比较,模型组大鼠的体质量均下降(P<0.01)、抑郁样行为学特征明显(P<0.01,P<0.05),血浆GLU含量升高、5-HT含量降低(均P<0.01)。CUMS配合孤养可致大鼠出现抑郁行为;与抑郁模型组比较,配合高脂饲料喂养抑郁模型组大鼠,除游泳不动时间外,抑郁行为学特征明显(P<0.05,P<0.01),GLU无明显变化,5-HT升高(P<0.05);两个给药组与模型组比较,给药组能显著对抗抑郁刺激,大鼠的体质量增加、行为学特征改善(均P<0.05,P<0.01),GLU的含量降低、5-HT升高(均P<0.05)。结论高脂饲料对抑郁大鼠的神经递质GLU无明显影响,但能升高5-HT加重老鼠的抑郁行为,百合知母汤提取物有很好的抗抑郁作用,其机制可能与调节抑郁症大鼠的神经递质,降低GLU、升高5-HT有关。
Objective To observe the antidepressant effect of extracts from Baihe Zhimu Decoction(BZD) on the depressive rat model fed by high- fat diet. Methods Fifty male SD rats were randomly divided into 5 groups,normal group,depressive model group,depressive model treated with BZD extract group,high- fat feeding depressive model group, and high- fat feeding depressive model treated with BZD extract group. The depressive model was induced by chronic unpredictable mild stress(CUMS) and solitary feeding. The modeling time lasted for 20 weeks. During the modeling,rat body weight was monitored,and behavioral characteristics of the rats were evaluated by the open field test and the forced swim test(FST). The contents of plasma glutamate(GLU) and 5- hydroxytryptamine(5- HT) were detected by using enzyme- linked immunosorbent assay(ELISA). Results Compared with the normal group,the model groups gained weight slowly,the behavioral characteristics were obvious,and plasma GLU content elevated and 5- HT decreased,the difference being statistically significant(P〈0.01).CUMS and solitary feeding could induce depression phenomenon in rats. Compared with the the depressive model group, high- fat feeding depressive model group had obvious behavioral characteristics(P〈0.01). The two medication groups showed an obvious effect on counteracting depression stress, decreasing body weight, improving rat behavior, reducing GLU content but increasing 5- HT content as compared the corresponding model group. Conclusion High- fat feeding has no effect on neurotransmitters,but can promote the development of depression. BZD extract has certain antidepressant, and the mechanism may be related to regulation of in- vivo neurotransmitter through reducing GLU and increasing 5- HT.
