大蒜素对阿尔茨海默病转基因小鼠脑内Tau蛋白表达的影响 被引量:4

摘要 本文应用免疫荧光技术和Western blot方法检测大蒜素对tau转基因小鼠脑内磷酸化tau蛋白表达的影响,并测定野生型小鼠、tau转基因小鼠及大蒜素治疗组三组小鼠脑内SOD活力和MDA含量,结果发现大蒜素处理之后的阿尔茨海默病(AD)模型小鼠SOD与AD组相比活性增加,MDA含量减少,tau转基因小鼠脑内所检测的Ser202、Ser404、Ser396、Thr181位点tau蛋白发生过度磷酸化改变,大蒜素治疗组这些过度磷酸化的tau蛋白明显减少。以上结果提示大蒜素可以通过其强大的抗氧化作用降低AD小鼠脑内氧化应激效应,从而使AD小鼠脑内tau蛋白磷酸化水平下降,因此大蒜素在AD的临床治疗方面具有重要的理论意义。 In this study, the effect of alliein on the expression of tau protein phosphorylated in the brain of transgenie mice was investigated by immunofluorescence and Western blot method. The SOD activity and MDA content in the brain of wild type,tau transgenie mice and the treatment group mice were detected. The results showed that the activity of superoxide dismutase(SOD) in the allicin administration mice brain was increased compared to that of AD model mice, and the content of MDA was reduced; Hyperphesphorylation of tan protein were changed at ser202, set404, ser396, Thrl81 site in the transgenic mice braln,but the hyperphosphorylation of tau protein was significantly reduced in the transgenic mice treated with allicin. These results indicated that allicin decreased oxidative stress effect in AD mice brain through its powerful antioxidant activity, and hence declined the level of tau protein phosphorylation in AD mice brain. These results suggested that allicin had potential value for the clinical treatment of AD.
