摘要
目的观察降脂酮对胰岛素抵抗大鼠肝病理损伤的保护作用。方法将15只大鼠随机分成2组,即正常对照组5只、双高加t BHP(HF+t BHP)组10只。正常对照组,正常饲料喂养,5-6周隔天腹腔注射生理盐水。HF+t BHP组,高脂高果糖饲料喂养,5-6周隔天腹腔注射t BHP。第6周末,将HF+t BHP组随机分成两组:模型组5只,降脂酮组5只。模型组,双高饮食+t BHP,生理盐水灌胃;降脂酮组,双高饮食+t BHP,降脂酮200 mg/kg灌胃。6周末,测定空腹血糖、血清胰岛素含量、胰岛素敏感指数。12周末,处死大鼠,腹主动脉取血,分离血清,测定大鼠ALT和AST。取肝脏作HE染色石蜡切片和制作电镜切片。结果与对照组比较,模型组空腹血糖升高(P<0.05),血清胰岛素含量降低(P<0.05),胰岛素敏感指数降低(P<0.05)。降脂酮治疗后,光镜下观察肝细胞排列整齐,肿胀减轻;电镜下观察肝脏线粒体肿胀减轻,内质网扩张不明显。大鼠血清中ALT和AST的活性显著降低(P<0.05)。结论降脂酮能够保护高脂高果糖饮食加腹腔注射t BHP诱导的胰岛素抵抗大鼠的肝损伤。
Objective To explore the role of Jiangzhitong in the pathological damage of the liver in rats with insulin resistance.Methods Fifteen rats were randomly divided into two groups: control group( n = 5) and high fat and high glucose plus t BHP group( HF + t BHP,n = 10). Control rats were fed with normal diet and injected with saline every other day from the fifth to the sixth week.HF + t BHP rats were given high-fat,high-fructose diet,and they were injected intraperitoneally with t BHP from the fifth to the sixth week. At the end of the sixth week,the HF + t BHP rats were randomly divided into two groups: model group( n = 5) and Jiangzhitong group( n = 5). The rats in model group were given high-fat,high-fructose diet and injected with t BHP from the seventh to the twelfth week,and the rats in Jiangzhitong group were given high-fat,high-fructose diet,Jiangzhitong 200 mg / kg orally and injected with t BHP. At the end of the sixth week,fasting blood glucose,serum fasting insulin levels,insulin sensitivity index were measured. At the end of the twelfth week,the rats were sacrificed and the livers were prepared for HE staining,oil red staining and electron microscopy slice. Results Compared with control group,the fasting blood glucose increased( P〈0. 05),the serum insulin level decreased( P〈0. 05),and the insulin sensitivity index decreased in model group( P〈0. 05). After treated with Jiangzhitong,liver cells were arranged neatly,and the swelling of liver cells was relieved. The results of electron microscope showed that the swelling of the mitochondrial was relieved in liver cells in Jiangzhitong group,and the endoplasmic reticulum wasn't obvious. The ALT and AST in serum of rats were decreased in Jiangzhitong group( P〈0. 05). Conclusion Jiangzhitong can alleviate the liver injury in insulin resistance rats induced by high fructose,high fat and t BHP.
