摘要
目的观察肥胖及代谢综合征幼鼠肝X受体α(liver X receptor alpha,LXRα)的表达。方法 3周SD大鼠随机分为普通饮食组(ND组),高脂饮食组(FD组)和高脂高盐饮食组(FSD组)。测量体质量、体长、腹围、血压、血脂、内脏脂肪质量等,口服葡萄糖耐量试验及胰岛素释放试验评价血糖及胰岛细胞功能,计算胰岛素抵抗指数。取新鲜肝脏组织,测定LXRαmRNA的含量。结果 FD组肥胖相关指标及胰岛素抵抗指数显著高于ND组,差异具有统计学意义(P<0.05)。FSD组血压、血糖、胰岛素、胰岛素抵抗指数、血脂等代谢综合征相关指标均显著高于ND组,差异具有统计学意义(P<0.05)。FD组及FSD组LXRαmRNA的表达显著高于ND组,差异具有统计学意义(P<0.01)。FD组及FSD组LXRαmRNA的表达差异无统计学意义(P>0.05)。结论肥胖及代谢综合征幼鼠LXRαmRNA表达增加,提示LXRα参与了胰岛素抵抗,糖脂代谢紊乱的病生理过程。LXRαmRNA可以作为肥胖及代谢综合征的检测指标。
Objective To investigate the expression of liver X receptor alpha( LXRα) mRNA in young rats with obesity and metabolic syndrome. Methods The 3-week-old SD young rats were divided into three groups: routine diet group( normal control group,n =12),high fat diet group( FD group,n = 14),and high fat and high salt diet group( FSD group,n = 14). The body weight,length,abdominal circumference,serum lipids and blood pressure,visceral fat weight were measured. Oral glucose tolerance test and insulin release test were performed to evaluate blood glucose and pancreatic beta cell function,and the insulin resistance index was calculated.LXRα mRNA was detected in flesh liver samples. Results In FD group,the obesity-related parameters and the insulin resistance index were significantly higher than that in normal control group( P〈0. 05). In FSD group,the metabolic syndrome-related parameters were significantly higher than that in normal control group( P〈0. 05). The LXRα mRNA expression in FD group and FSD group was higher than that in normal control group( P〈0. 01),but there was no significant difference between FD group and FSD group( P〈0. 05). Conclusion The expression of LXRα mRNA in young rats with obesity and metabolic syndrome is increased,suggesting that LXR is involved in the process of insulin resistance and glucose and lipid metabolism disorders. LXRα can be used as an indicator of obesity and metabolic syndrome.
