白藜芦醇对荷卵巢癌大鼠免疫功能的影响 被引量:7

摘要 目的研究白藜芦醇(resveratrol,Res)对荷卵巢癌大鼠免疫功能的影响。方法通过右腋窝皮下注射接种卵巢癌NUTU-19细胞悬液的方法制备荷卵巢癌大鼠模型,取50只模型大鼠随机分为:模型对照组、白藜芦醇(200、100、50mg/kg)治疗组和顺铂2mg/kg治疗组(阳性对照);白藜芦醇各治疗组隔天灌胃给药1次,顺铂组隔天腹腔注射给药1次,共给药治疗5次。治疗完成后,观察比较各组大鼠一般状态及皮下肿瘤生长情况;处死动物后剥取肿瘤组织,称量瘤重量并计算抑瘤率;通过四甲基偶氮唑盐(MTT)染色法测定各组大鼠脾淋巴细胞转化率;通过酶联免疫吸附法(ELISA)测定各组大鼠血清中白细胞介素-2(interleukin-2,IL-2)和肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)含量;通过流式细胞仪检测各组大鼠血中T淋巴细胞亚群(CD4^+、CD8^+)百分率,并计算CD4^+/CD8^+比值;通过苏木精-伊红(HE)染色观察肿瘤组织病理性形态学变化。结果与模型对照组比较,白藜芦醇治疗组大鼠皮毛松散、饮食差状况减轻,瘤体减小、活动度差、硬度差,以200mg/kg治疗组效果最为显著;白藜芦醇(200、100mg/kg)治疗组肿瘤重量显著减轻且抑瘤率显著升高(P<0.05);脾淋巴细胞转化率显著升高(P<0.05);血清中IL-2和TNF-α含量均显著降低(P<0.05);白藜芦醇(200、100mg/kg)治疗组血液中CD4^+百分率显著升高(P<0.05),白藜芦醇200mg/kg治疗组CD8^+百分率显著升高,CD4^+/CD8^+比值显著升高(P<0.05);白藜芦醇治疗组肿瘤组织呈现片状坏死、瘤细胞皱缩等病理性形态学变化,以200mg/kg治疗组最为明显。结论白藜芦醇能够显著提高卵巢癌大鼠免疫功能,作用机制可能与其能够有效降低炎性因子(IL-2、TNF-α)水平,提高CD4^+、CD8^+百分率及CD4^+/CD8^+比值有关。 Objective To investigate the effects of resveratrol(Res) on immune function of ovarian cancer rats. Methods Fifty ovarian cancer rat models made by injecting ovarian cancer cell line NUTU - 19 were divided randomly into five groups: model control group, Res(200, 100, 50mg/kg) groups and Cisplatin 2mg/kg group, the Res were given by intragastric administration (ig) and the Cisplatin was given by intraperitoneal injection for 5 times, once every two days. The general states was observed, the tumor weight were detected, and inhibition rate was calculated. The spleen lymphocyte transformation rate was detected by MTT; and the content of interlenkin- 2 (IL- 2) and Tumor necrosis factor- α(TNF- α) in serum were detected. The present of CD4^+ , CD8^+ in blood were detected, and the ratio of CD4^+/CD8^+ ; the histopathological changes of tumor tissue was observed by HE staining. Results Compared with model control group, the rats in Res groups appeared fur loose, poor diet, significantly reduced the tumor, poor activity, the hardness difference. The tumor weight were significantly decreased and the inhibition rate were significantly increased(P 〈 0.05 ). The spleen lymphocyte transformation rate of Res(200, 100mg/kg) groups were significantly increased( P 〈 0. 05). The content of IL - 2 and TNF -α in serum were significantly decreased( P 〈 0.05 ). The CD4^+ present in blood were significantly increased( P 〈 0. 05 ). The CD8^+ present in blood of Res 200mg/kg group was significantly increased (P 〈 0.05). The ratio of CD4^+/CD8^+ of Res (200, 100mg/kg) groups were significantly increased (P 〈 0.05). Necrosis presentation,tumor cell shrinkage and other pathological morphological changes appeared in Res (200, 100mg/kg) groups. Conclusion Res could effectively improve the immune function of ovarian cancer rat, which perhaps related to its effects of reducing the inflammatory cytokines (IL- 2, TNF -α) levels and increasing CD4^+, CD8^+ percentages and CD4^+/ CD8^+ ratio.
