摘要
目的:构建人皮肤慢性光老化成纤维细胞模型,筛查与光老化相关的环状RNA(circRNA)。方法:分离培养成纤维细胞,利用UVA多次照射,制作人皮肤慢性光老化成纤维细胞模型,采用β-半乳糖苷酶染色及Western印迹法检验p21蛋白验证,以给予与照光时长等同的避光处理的成纤维细胞作为对照。采用RNAseq高通量转录组测序技术检测光老化成纤维细胞中circRNA表达,并分析其差异表达的circRNA。结果:与对照组比较,光老化模型组成纤维细胞中表达显著上调的circRNAs共有7个(P<0.05);表达显著下调的circRNAs共有4个(P<0.05)。经生物信息学分析,差异表达的circRNA可能与胶原蛋白合成等基因有关。结论:光老化模型组成纤维细胞存在多个差异表达的circRNAs,说明circRNA可能参与光老化的基因调控。
Objective:To screen and identify the differentially expressed circRNA in photoaged model of human fibroblasts. Methods: Human dermal fibroblasts (HDFs) at early passages were exposed to a series of sub-cytotoxic dose of UVA. The senescent phenotypes were detected by the expression of ^-galaetosidase and p21 protein. Circular RNAs expression were identified in pho- toaged HDFs through high-throughput deep sequencing coupled with analysis of massive transcrip- tional data. Results:Sequencing result revealed that 11 eircRNAs were significantly altered in the photoaged model (P 〈 0. 05 ) compared with the control group. Among them, 7 were significantly up-regulated, and the other 4 were down-regulated. Bioinformaties analysis revealed that those cir- cRNAs may involve in encoding the chains of collagen. Conclusion:This study results confirmed some circRNAs were differentially expressed in photoaged HDFs model, which may participate in the process of gene regulation of photoaging.
