GSTP1基因表达及甲基化与前列腺癌复发风险研究 被引量:2

摘要 目的探究谷胱苷肽S转移酶P1(glutathione S transferase pi,GSTP1)基因表达及甲基化水平与前列腺癌复发风险的关系。方法利用癌症基因图谱(the cancer genome atlas,TCGA)数据库,分析前列腺癌组织中GSTP1基因表达及甲基化水平,采用t检验、单因素及多因素logistic回归探讨GSTP1基因与前列腺癌复发的关联性。结果前列腺癌GSTP1基因表达量与前列腺癌的复发显著相关,校正后的相对危险度(odds ratio,OR)为0.63,P=0.039;此外,相对于非复发组,GSTP1基因启动子区有5个CpG位点在复发组中均呈现高甲基化状态,且能显著增加前列腺癌复发风险。结论 GSTP1基因表达与前列腺癌复发显著相关,可能是通过GSTP1启动子区CpG位点的高甲基化进而减低GSTP1基因的表达,从而影响前列腺癌的复发。 Objective To investigate the association of recurrence and expression and methylation of GSTP1 in prostate cancer. Methods Data of expression and methylation of GSTP1 was downloaded from TCGA database and association study was performed using T test,univariate and multivariate logistic regression. Results The expression of GSTP1 in recurrence group was significantly lower than non-recurrence group.( adjusted OR = 0. 63,P = 0. 039) A total of five CpG loci significantly increased the risk of prostate cancer recurrence,with consistent hypermethylation in recurrence group( P〈0. 05). Conclusions Down-regulation of GSTP1 gene expression in prostate cancer recurrence group may be caused by hypermethylation in promoter region of GSTP1.
