Establishment of a rat model of accelerating future liver remnant regeneration

摘要 目的建立一种加速剩余肝脏(future liver remnaut volume,FLR)再生的大鼠模型。方法雄性SD大鼠96只,应用计算机产生随机化数字进行完全随机化分组,分为3组,每组32只,即:肝脏离断+门静脉结扎组(LP+PVL组)、门静脉结扎组(PVL组)和假手术组(Sham组)。评估术后24 h、72 h、7 d和10 d时各组FLR再生率、FLR重量/总肝重、肝功能变化和肝脏组织学改变。结果术后24 h、72 h、7 d和10 d时,LP+PVL组的FLR再生率分别为(29.97±8.86)%、(76.67±9.58)%、(108.61±2.65)%和(117.21±2.66)%,明显高于PVL组(P<0.05),FLR重量/总肝重也显示类似变化。LP+PVL组和PVL组的术后肝功能和剩余肝脏组织学检查均显示有一定损害,且以LP+PVL组明显;但两组均于术后10d恢复正常。结论肝脏离断加门静脉结扎能有效促进剩余肝脏再生,术后10 d FLR再生率可达117%、FLR重量/总肝重可达60%。 Objective To establish a rat model of accelerating future liver remnant( FLR) regeneration.Methods 96 male SD rats were randomly divided into the following three groups( 32 rats each group) : liver partition + portal vein ligation( LP + PVL group),portal vein ligation( PVL group) and sham-operated group( Sham group). The changes of the regeneration rate of FLR,FLR weight/total liver weight,the liver functions,and the liver histopathology were measured after 24 h,72 h,7 d and 10 d in each group. Results After 24 h,72 h,and 7 d and 10 d,the regeneration rate of FLR were( 29. 97 ± 8. 86) % 、( 76. 67 ± 9. 58) % 、( 108. 61 ±2. 65) % and( 117. 21 ± 2. 66) % in LP + PVL group,significantly higher than the PVL group( P〈0. 05),FLR weight / total liver weight also showed similar changes. The liver functions and the liver histopathology all showed there were some damage after operation in LP + PVL group and PVL group,and LP + PVL group was significant. However,both groups returned to normal after 10 d. Conclusions Liver partiton and portal vein ligation can effectively promote future liver remnant regeneration. The regeneration rate of FLR can be up to117%,and FLR weight / total liver weight can be up to 60% after 10 d.
