瑞舒伐他汀对脑缺血再灌注大鼠白细胞介素-23/白细胞介素-17炎症轴的影响 被引量:5

Effects of rosuvastatin on interleukin-23/interleukin-17 inflammatory axis in rats with focal cerebral ischemia-reperfusion

摘要 目的研究瑞舒伐他汀对脑缺血再灌注损伤大鼠白细胞介素(IL)-23和IL-17表达的影响。方法将24只SD大鼠随机分为假手术组、模型组和实验组,每组8只。模型组和实验组按改良Zea-Longa线栓法制备大鼠大脑中动脉栓塞模型,血流阻断1 h后拔出线栓,形成再灌注。假手术组大鼠接受相似手术处理,但是不插入线栓。实验组在模型制作前用瑞舒伐他汀5 mg·kg^(-1)·d^(-1)连续灌胃10 d,qd;假手术组和模型组予以正常喂养。再灌注12 h后,进行神经功能评分,处死大鼠断头取脑,用半定量聚合酶链式反应法和Western blot法分别检测脑组织中IL-23和IL-17的表达水平。结果假手术组无神经功能缺损,IL-23 mRNA和蛋白及IL-17 mRNA和蛋白呈低水平表达。脑缺血再灌注损伤后,与假手术组比较,模型组神经功能评分升高,IL-23 mRNA和蛋白及IL-17mRNA和蛋白表达均明显增高(P<0.05)。与模型组比较,实验组神经功能评分降低,IL-23 mRNA和蛋白及IL-17 mRNA和蛋白表达均明显减少(P<0.05)。结论瑞舒伐他汀可能通过抑制IL-23/IL-17炎症轴发挥脑缺血保护作用。 Objective To explore the effects of rosuvastatin on the expression of interleukin( IL)-23 and IL-17 in rats with focal cerebral ischemia-reperfusion.Methods Rats were randomly assigned into sham-operation group,model group and test group.The model rats with middle cerebral artery occlusion were prepared by the modified Zea-Longa suture method.After vascular occlusion 1 h,remove the suture to form reperfusion.Rats in sham-operation group received similar surgical treatment,but didn 't insert the suture.The rats in the test group were lavaged with rosuvastatin for 10 consecutive days before the model setting-up.The rats in the sham-operation group and model group were fed normally.At 12 h after reperfusion,scores of neurological deficit were estimated.The levels of IL-23 and IL-17 were measured by real-time polymerase chain reaction and Western blotting.Results The rats in the sham-operation group had no neurological deficit,the levels of IL-23 mRNA,protein and IL-17 mRNA,protein appeared lower expression.Compared with sham-operation group,the scores of neurological deficit and the levels of IL-23 mRNA,protein and IL-17 mRNA,protein increased significantly in rats after focal cerebralischemia-reperfusion( P〈0.05).Compared with model group,rosuvastatin significantly decreased the scores of neurological deficit and the levels of IL-23 mRNA,protein and IL-17 mRNA,protein( P〈0.05).Conclusion Rosuvastatin may protect the brain injury through inhibiting the expression of IL-23 /IL-17 inflammatory axis.
