红木生育三烯酚对结肠癌小鼠免疫相关细胞与细胞因子的作用 被引量:1

EFFECTS OF ANNATTO TOCOTRIENOL ON IMMNUE CELLS AND CYTOKINES IN COLORECTAL CANCER MICE

摘要 目的研究红木生育三烯酚(δ-T_3)对结肠癌小鼠CD4^+T细胞、CD8^+T细胞、白介素(IL)-6、干扰素(IFN)-γ的调节作用。方法 75只CD1-ICR小鼠分为结肠癌造模组和阴性对照两大组,结肠癌造模组再分为肿瘤模型对照组和三个δ-生育三烯酚剂量组(分别5、10、20mg/kg·bw),每组15只。采用氧化偶氮甲烷(AOM)和葡聚糖硫酸钠(DSS)诱导结肠癌模型。采用全自动五分类血球仪毛细管模式检测动物血液学指标,流式细胞仪检测外周血T淋巴细胞亚型,ELISA法检测血清白细胞介素(IL)-6、干扰素(IFN)-γ水平。结果与空白对照组比较,结肠癌造模组ICR小鼠白细胞计数降低(P<0.05)。与肿瘤模型对照组比较,中、高剂量生育三烯酚组均能显著提高CD4^+/CD8^+比值和血清中IFN-γ的水平,降低IL-6水平(P<0.05)。结论δ-生育三烯酚可增强结肠癌小鼠的免疫功能,这可能与其提高CD4^+/CD8^+比值,提高IFN-γ水平,及降低IL-6水平相关。 Objective To study the effect of annatto tocotrienol(δ-T_3) on CD4+, CD8+T lymphocytes, interleukin(IL)-6 and interferon(IFN)-γ in colorectal cancer mice. Methods Seventy-five CD1-ICR mice were randomly divided into control group, colorectal cancer model control group, low(5mg/kg·bw), middle(10mg/kg·bw) and high dose δ-T3 treatment groups(20mg/kg·bw) with equal size. Azoxymethane(AOM) and dextran sulfate sodium(DSS) were used to induce colorectal cancer model. Automatic classification blood capillary pattern was used to detect hematological indexes. T lymphocyte subsets were measured by flow cytometry. IL-6 and IFN-γ were measured by ELISA. Results Compared with control group, leukocyte count decreased significantly in colorectal cancer model group(P〈0.05). Compared with model control group, the ratio of CD4+/CD8+ and the level of IFN-γ increased significantly in high and middle dose tocotrienol groups, while the level of IL-6 decreased in those groups(P〈0.05). Conclusion δ-tocotrienol could enhance immune function in colorectal cancer mice, which might be correlated with its ability of increasing the ratio of CD4+/CD8+ and the level of IFN-γ, and decreasing the level of IL-6.
