针药结合对糖尿病胃轻瘫小鼠胃窦酪氨酸激酶受体及干细胞因子的影响

Effect of Acupuncture Combined with Chinese Medicine on Diabetic Gastroparesis Antrum Tyrosine Kinase Receptor and Stem Cells Factor

目的:研究针药结合对糖尿病胃轻瘫小鼠胃窦酪氨酸激酶受体(c-Kit)及干细胞因子(SCF)的影响。方法:80只昆明小鼠除正常组之外,均一次性腹腔注射链脲佐菌素及高糖高脂喂养,将诱导成功的糖尿病胃轻瘫小鼠模型随机分为模型组、针药组、中药组和西药组;针药组采取后三里、中脘电针合并中药方糖瘫消灌胃(0.026 m L/g,1次/d);中药组仅予以糖瘫消;西药组予以多潘立酮和维生素B_6混合液。治疗结束后,采用Western blot法检测胃窦c-Kit蛋白的变化、RT-PCR法测定胃窦平滑肌组织SCF mRNA的表达。结果:与正常组比较,模型组c-Kit蛋白和SCF mRNA表达显著下调(P<0.01);治疗后,与模型组比较,仅有针药组c-Kit蛋白和SCF mRNA表达显著增加(P<0.01);治疗组间,与针药组比较,中药组和西药组的c-Kit蛋白及SCF mRNA表达均显著下调(P<0.05或P<0.01)。结论:针药结合对糖尿病胃轻瘫模型小鼠有较好的治疗效果,其机理可能与促进c-Kit蛋白及SCF基因的表达,部分恢复c-Kit、SCF的损伤及修复SCF-Kit信号途径有关。

Objective： To observe the effect of acupuncture combined with Chinese medicine on diabetic gastroparesis antrum tyrosine kinase receptor（ c- Kit） and stem cells factor（ SCF）. Methods： Eighty Kunming male rats were equally randomized into five groups： normal group,diabetic gastroparesis model rats group（ model group）,combination of acupuncture and Chinese medicine group（ ACM group）,Chinese medicine group（ CM group） and Western medicine group（ WM group）. The Western blot was conducted to detect gastric tissue c- Kit protein expression levels and SCF mRNA to determine the expression of gastric tissue.Results： Compared with the normal group,the indexes of c- Kit protein expression levels and SCF mRNA in model group were significantly reduced（ P 0. 01）. After treatment,compared with model group,only ACM group＇s expressions of c- Kit protein and SCF mRNA increased significantly（ P 0. 01）. Compared with ACM group,the indexes of c- Kit protein expression levels and SCF mRNA in CM group and WM group were significantly lower（ P 0. 05,P 0. 01）. Conclusion： Acupuncture combined with Chinese medicine has better therapeutic effects for diabetic gastroparesis rats and the mechanism may be related to promoting c-Kit protein and SCF mRNA expression levels,which may contribute to its effect on recovery of form and function of c- Kit protein and SCF and then repairing the signal channel of SCF- Kit.
