填精补脑方对老年痴呆大鼠学习记忆及海马单胺类神经递质的影响 被引量:1

Effect of replenishing essence and tonifying brain prescription on learning memory and monoamine neurotransmitter in hippocampus in rats with Alzheimer's disease

摘要 目的观察填精补脑方对老年痴呆大鼠学习记忆及海马单胺类神经递质的影响。方法 2014年5—8月将40只健康成年SD大鼠随机分为对照组、模型组、脑复康组、填精补脑方组各10只。双侧海马注射Aβ25-35构建老年痴呆大鼠模型,各组给予相应药物灌胃治疗20 d。用药结束后,测定大鼠的学习记忆能力并检测海马组织的去甲肾上腺素(noradrenaline,NE)、多巴胺(dopamine,DA)、5-羟色胺(5-hydroxytryptamine,5-HT)。计量资料比较采用重复测量资料方差分析和单因素方差分析,P<0.05为差异有统计学意义。结果与对照组[(36.48±4.67)、(29.27±4.25)、(22.84±2.71)、(16.23±1.58)s]相比,第2、3、4、5天模型组大鼠的逃避潜伏期(escape latency,EL)[(48.53±5.17)、(42.14±3.87)、(38.63±3.56)、(35.62±3.26)s]明显延长,脑康复组和填精补脑方组大鼠的EL[(39.16±3.23)、(34.56±3.28)、(28.25±2.49)、(23.46±2.31)s与(39.87±3.67)、(34.98±3.59)、(28.67±2.58)、(23.87±2.37)s]明显缩短,差异均有统计学意义(均P<0.05)。与对照组[(0.596±0.076)、(0.785±0.069)、(0.687±0.057)μg/g]比较,模型组大鼠海马中的NE、DA、5-HT[(0.341±0.035)、(0.562±0.076)、(0.457±0.079)μg/g]明显降低,差异均有统计学意义(均P<0.05);脑复康组和填精补脑方组大鼠海马中的NE、DA、5-HT[(0.497±0.065)、(0.693±0.057)、(0.658±0.056)μg/g与(0.493±0.059)、(0.689±0.078)、(0.654±0.048)μg/g]明显增加,差异均有统计学意义(均P<0.05)。结论填精补脑方能提高老年痴呆大鼠的学习记忆能力,改善海马中单胺类神经递质代谢。 Objective To study the effect of replenishing essence and tonifying brain prescription on learning and memory and monoamine neurotransmitter in hippocampus in rats with Alzheimer's disease.Methods Healthy adult SD rats were randomly divided into 4 groups: control group,model group,piracetam group,replenishing essence and tonifying brain prescription group from May to August in 2014.There were 10 rats in each group.The model of Alzheimer's disease was established with microinjection Aβ25-35 on bilateral hippocampus.All rats were treated with corresponding drugs respectively lasting 20 days.Then,the learning and memory ability of rats were examined.and the contents of noradrenaline(NE),dopamine(DA) and 5-hydroxytryptamine(5-HT)on hippocampus were measured.Measurement data was processed by repeated measures analysis of variance and one-way variance analysis,P0.05 was considered statistically significant.Results Compared with control group [(36.48±4.67),(29.27±4.25),(22.84±2.71),(16.23±1.58)s],the escape latency(EL) of model group in 2nd,3rd,4th,5th day [(48.53±5.17),(42.14±3.87),(38.63±3.56),(35.62±3.26)s] prolonged significantly(all P0.05).The EL of piracetam group,replenishing essence and tonifying brain prescription group [(39.16±3.23),(34.56±3.28),(28.25±2.49),(23.46±2.31)s and(39.87±3.67),(34.98±3.59),(28.67±2.58),(23.87±2.37)s] shortened significantly(all P0.05).Compared with control group[(0.596±0.076),(0.785±0.069),(0.687±0.057)μg/g],NE,DA,5-HT of model group [(0.341±0.035),(0.562±0.076),(0.457±0.079)μg/g] decreased significantly(all P0.05).NE,DA,5-HT of piracetam group,replenishing essence and tonifying brain prescription group [(0.497±0.065),(0.693±0.057),(0.658±0.056)μg/g and(0.493±0.059),(0.689±0.078),(0.654±0.048)μg/g] increased increased significantly(all P0.05).Conclusion Replenishing essence and tonifying brain prescription could raise the learning and memory ability of Alzheimer's disease rats and improve the metabolism of monoamine neurotransmitter in hippocampus.
