摘要
[目的]研究阻断PI3K/Akt信号通路对布鲁氏菌介导的细胞凋亡的影响。[方法]用抑制剂LY294002(LY)作用于细胞1 h,然后将布鲁氏菌16 M侵染细胞,应用实时定量PCR检测16 M对巨噬细胞内凋亡相关基因Caspase-3活性和Bax mRNA表达的影响。[结果]阻断PI3K/Akt信号通路可显著提高16 M介导的caspase-3活性和Bax mRNA水平。[结论]PI3K/Akt信号通路可调控布鲁氏菌介导的细胞凋亡。
[Objective]To detect the changes of Brucella mediated apoptosis genes regulated by PI3 K/Akt signal pathway. [Method] Inhibitor LY294002( LY) was applied on cells for 1 h. Brucella 16 M was applied to infect the cells. Real time quantitative PCR was used to detect the effects of 16 M on macrophage apoptosis related gene Caspase-3 activity and Bax mRNA expression.[Result] Blocking PI3 K/Akt signal pathway could significantly enhance the Caspase-3 activity and Bax mRNA level mediated by 16 M. [Conclusion] PI3 K/Akt signal pathway can regulate the Brucella mediated apoptosis.
