摘要
目的:分离和识别一株抗真菌毛霉菌,并测试其抗菌活性。方法:采用稀释平板涂布法分离土壤中真菌;以杯碟法初步测定发酵液抗真菌活性;通过菌落及孢子进行形态学鉴定;以薄层色谱-生物自显影技术分离检测抗菌活性成分。结果:从土壤中分离出一株对白色念珠菌具有较强抑制活性的菌株F1,抑菌圈直径为20.5 mm,形态学初步鉴定为毛霉;该毛霉发酵液乙酸乙酯相在甲醇∶三氯甲烷(1∶4)展开系统,结果显示有两个组分对白色念珠菌具有抑制活性,其Rf值分别为0.15和0.6。结论:来自土壤的毛霉发酵液乙酸乙酯相具有抑制白色念珠菌的活性组分。
Objectiyve: To isolate and identify an antifungal mucor,and test its antibacterial activit. Methods: Dilution spread plate method was used to isolate the fungi from soils,and plate method was applied to determining the antifungal activity of fermentation broth. The strain was identified in compliance with the morphology and spore configuration. Thin-layer chromatography-bioautography assay was performed to detect antibacterial compound in the isolated fungi. Results: One strain mucor capable of inhibiting the growth of C. albicans with an inhibition diameter of 20. 5 mm was obtained by initial morphological identification. Two antimicrobial components were extracted in ethyl acetate with expand system of methanol and chloroform( ratio: 1 ∶ 4),and showed inhibitory activity against C. albicans. The retardation factor( Rf) value was 0. 15 and 0. 6,respectively. Conclusion: The mucor,an extraction with ethyl acetate,yields active compound against C. albicans.
