基于核磁共振技术的益肾喘宁汤对支气管哮喘肾气虚证模型大鼠血清代谢组学的影响 被引量:17

Effect of Yishen Chuanning Decoction(益肾喘宁汤) on Serum Metabonomics in Rats Model with Bronchial Asthma of Kidney-qi Deficiency Syndrome: Based on Nuclear Magnetic Resonance Technics

摘要 目的利用核磁共振技术探讨益肾喘宁汤治疗支气管哮喘肾气虚证的可能作用机制。方法将42只Wistar大鼠随机分成正常对照组、模型组、中药组各14只,除正常对照组外,其余各组大鼠均采用卵蛋白联合游泳力竭法、恐伤肾双因素复合法建立支气管哮喘肾气虚证模型。实验第15天开始中药组大鼠给予益肾喘宁汤2.0 g/(kg·d)灌胃,正常对照组和模型组给予等量的生理盐水灌胃。各组大鼠连续灌胃15天后腹主动脉采血分离血清,进行氢谱核磁共振检测,最终筛选并鉴定出各组大鼠血清中的差异代谢物。结果共鉴定出20种差异具有统计学意义的血清代谢物,相对于正常对照组,模型组大鼠血清中异亮氨酸、谷氨酰胺含量显著升高,乳酸含量显著降低。相对于模型组,中药组大鼠血清中乳酸含量显著升高,脂类(R-CH_3)、脂类(R-CH_2)、脂类(CH2CH_2CO)、脂类(CH_2C=C)、脂类(C=CCH_2C=C)以及不饱和脂肪酸的含量降低(P<0.05)。结论益肾喘宁汤治疗支气管哮喘肾气虚证的可能主要机制为促进机体脂类代谢,进而改善机体能量代谢缺乏状态。 Objective To explore the possible mechanism of Yishen Chuanning Decoction( 益肾喘宁汤) in treating bronchial asthma of kidney-qi deficiency syndrome using nuclear magnetic resonance. Methods Randomly divided 42 Wistar rats into normal control group,model group and the Chinese medicine group,with 14 rats in each.Except for the normal control group,rats were treated with exhausting swimming and great fright impairing the kidney to establish bronchial asthma of kidney-qi deficiency syndrome model. The Chinese medicine group was given Yishen Chuanning Decoction 2. 0 g /( kg·d) for gavage from the 15 th day of experiment. The normal control group and the model group were given equal dose saline for gavage. Collected blood from aorta abdominalis and separated serum of rats in each group after 15 days of continuous gavage. Conducted H-nuclear magnetic resonance examination. Finally selected and authenticated different metabolite of rats in different groups. Results Totally authenticated 20 kinds of serum metabolites whose difference had statistic significance. Compared to those in the normal control group,isoleucine and glutamine levels remarkably increased while that of the lactic acid remarkably decreased in the model group.Compared with the model group,lactic acid level in the Chinese medicine group remarkably increased,while the lipid( R-CH_3),lipid( R-CH_2),lipid( CH_2CH_2CO),lipid( CH_2 C = C),lipid( C = CCH_2 C = C) and unsaturated fatty acid levels decreased( P〈0. 05). Conclusion The possible functional mechanism of Yishen Chuanning Decoction( 益肾喘宁汤) in treating bronchial asthma of kidney-qi deficiency syndrome is to promote body lipid metabolism and improve energy metabolism deficiency of the body.
