摘要
采用去卵巢骨质疏松大鼠模型,观察跑台运动、全身垂直振动、金雀异黄酮、雌激素和氯化锂5种干预疗法对去卵巢骨质疏松大鼠骨髓脂肪细胞数目和过氧化物酶体增殖激活物受体γ2(PPARγ2)蛋白表达水平的影响。方法:将80只3月龄雌性SD大鼠按体重分层后随机分为假手术组和去卵巢组,去卵巢10周后,将去卵巢组大鼠按体重分层后又随机分为去卵巢组、雌激素组、金雀异黄酮组、氯化锂组、跑台运动组和全身垂直振动组。去卵巢第11周时,不同组别的大鼠开始进行不同干预处理。干预8周后,用HE染色法观察左胫骨组织形态学和骨髓脂肪细胞空泡数目的变化;用Western blotting检测右胫骨近端骨组织PPARγ2蛋白表达水平的变化。结果:经雌激素、金雀异黄酮、跑台运动和氯化锂干预后,去卵巢骨质疏松大鼠胫骨松质骨骨小梁数目显著增加,脂肪空泡数目和PPARγ2蛋白表达水平显著下降,而经全身垂直振动干预后,去卵巢骨质疏松大鼠胫骨松质骨骨小梁数目显著增加,脂肪空泡数量显著减少,但PPARγ2蛋白表达水平无显著变化。结论:雌激素、金雀异黄酮、跑台运动和氯化锂可能是通过下调去卵巢骨质疏松大鼠骨组织PPARγ2蛋白的表达水平而抑制去卵巢骨质疏松大鼠骨髓细胞向脂肪细胞的分化。
The aim of this study was to investigate the effects of five different intervention methods on the bone histology and the protein expression of peroxisome-proliferator-activated receptor-gamma2(PPARγ2)in the ovariectomized osteoporosis rats.Methods:Eighty healthy female SD rats,aged 3months,were randomly divided into the following two groups by body weight:sham-operation(Sham)and ovariectomized(OVX).After ten weeks,the OVX group rats were randomly divided into the following six groups by body weight:OVX;17β-estradiol(E2);Genisteine(G);treadmill exercise(TE);Lithium chloride(LiCL);Whole-body vertical vibration(WBVV).Then the rats began to be treated with different intervention methods.At the end of 8weeks intervention,during 36-48 hours,the tibia was isolated.The morphological changes of the left tibia were observed by HE staining and the protein expression of PPARγ2in right tibia were detected by western blot.Results:After treated with E2,treadmill exercise,whole-body vertical vibration,genistein and LiCL,the number of trabecular bone were significantly increased and the number of bone marrow adipocytes were significantly decreased than that of in OVX group.After treated with E2,treadmill exercise,genistein and LiCL,the protein expression of PPARγ2were significantly lower than that of in OVX group.However,whole-body vertical vibration treatment had no significant difference.Conclusions:Treadmill exercise,E2,genistein and LiCL could inhibit the differentiation of bone marrow stromal cells to fat cells by inhibiting the protein expression of PPARγ2in bones of OVX osteoporosis rats.
