益气活血中药对脂多糖致急性肺损伤大鼠的防治作用 被引量:5

摘要 目的观察益气活血中药对脂多糖(lipopolysaccharide,LPS)致急性肺损伤(acute lung injury,ALI)大鼠的防治作用,并初步探讨其作用机制。方法 SD雄性大鼠40只,随机分成正常对照组、模型对照组、地塞米松组、益气活血中药治疗组、益气活血中药防治结合组,每组8只;除正常对照组行气管内滴注生理盐水,其余各组均滴注LPS(5 mg/kg)制备ALI大鼠模型;造模前后各3天干预组予以2 m L相应药物(益气活血中药0.59 g/m L;地塞米松5 mg/kg),其余组予以等量的生理盐水灌胃。观察大鼠的呼吸、精神等一般活动;于末次灌胃给药2小时后处死大鼠,测定大鼠肺湿重/干重(W/D)比值,支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)中总蛋白含量,及光镜下观察肺组织病理学改变。结果 LPS造模组大鼠肺W/D比值、BALF中蛋白浓度及肺组织中肿瘤坏死因子(tumornecrosis factor-α,TNF-α)、白细胞介素(interleukin,IL)-1β、IL-6含量均高于正常对照组(P<0.01),且模型对照组高于地塞米松组和中药治疗组、中药防治结合组(P<0.05),治疗组高于防治组(P<0.05,P<0.01);地塞米松组W/D比值及肺组织中TNF-α、IL-1β含量与中药防治组无明显差异(P>0.05)。结论益气活血中药能够降低LPS致ALI大鼠BALF中蛋白含量及肺组织中炎症因子的含量,减轻肺水肿和肺组织的病理损伤,起到防治急性肺损伤的作用。 Objective To observe the preventive effect of Yiqi Huoxue recipe on acute lung injury( ALI) induced by lipopolysaccharide( LPS),and explore its mechanism. Methods Forty healthy male SD rats were randomly divided into normal control group,model control group,dexamethasone group,herbs treatment group,herbs prevention group,eight rats in each group. ALI models were established by intratracheal instillation of LPS( 5 mg / kg) while normal control group was instilled normal saline. 3 days before and after making the model,the drug( 0. 59 g / m L herbs or dexamethasone for 5mg / kg) or same amount of normal saline was given by gavage in each group,and observe their respiration,activity and so on.Rats were sacrificed after 2h of the last administration. Then the lung wet weight / dry weight( W / D) ratio and the protein content in bronchoalveolar lavage fluid( BALF) was measured,and the pathological changes in lung tissues. Results Compared with normal control group,LPS groups exhibited a higher level of the lung W / D weight ratio,the protein content in BALF and the content of TNF-α,IL-1β,IL-6 in lung tissue( P〈0. 01); compared with model control group,the indexes in dexamethasone group and herbs group were decreased( P〈0. 01); compared with herbs treatment group,the indexes in herbs prevention group were decreased( P〈0. 01 or P〈0. 05); the lung W / D weight ratio,the content of TNF-α,IL-1βbetween dexamethasone group and herbs prevention group were close( P〈0. 05). Conclusion Yiqi Huoxue recipe can reduce the contents of inflammatory cytokines and protein content in BALF of LPS induced ALI rats,reduce the pathological damage of lung tissue and pulmonary edema,have the effect of prevention and treatment of acute lung injury.
