锦灯笼宿萼提取物乙酸乙酯层改善东莨菪碱引起学习记忆损伤的研究

摘要 目的研究锦灯笼宿萼提取物乙酸乙酯层(PEE)对东莨菪碱引起的学习记忆损伤的作用效果及其作用机制。方法将锦灯笼宿萼用95%乙醇浸提、溶剂分配,得到乙酸乙酯层。以不同剂量[10、30、100 mg/(kg·d)]的PEE连续灌服小鼠14 d,腹腔注射2 mg/kg东莨菪碱造模。用新奇物体实验检测其对学习记忆的影响;测定海马区乙酰胆碱酯酶(ACh E)和超氧歧化酶(SOD)的活性以及丙二醛(MDA)的水平,研究其作用机制。结果新奇物体实验表明,10、30、100 mg/(kg·d)的PEE可以明显提高小鼠在新奇物体中的学习记忆能力;不同剂量的PEE可以抑制海马区ACh E活性并增加SOD的活性,降低MDA的水平。毒性研究表明,PEE对小鼠体重及脏器系数均无明显影响。结论 PEE通过抑制ACh E活性及增加抗氧化应激水平改善由东莨菪碱造成的学习记忆损伤,提示其具有潜在预防或治疗阿尔茨海默病的效果。 Objective To investigate the effects of PEE on scopolamine-induced learning and memory impairments and the action of mechanisms. Methods The Physalis alkekengi L.var.franchetii was extracted with 95% aqueous ethanol, the extraction was purified between water and ethyl acetate,and the ethyl acetate layer was used for the experiment.The mice were treated with PEE at doses of 10,30,100 mg/(kg.d) for 14 consecutive days,and then treated with scopolamine at the dose of 2 mg/kg.The cognitive-improving effect of PEE on amnesic mice induced by scopolamine was investigat- ed by assessing the novel object recognition test.In order to confirm the underlying mechanisms,activities of AChE and antioxidant enzymes were measured. Results Administration of PEE significantly restored scopolamine-induced memory impairments in the novel object recognition test.The increased AChE activities produced by scopolamine were signifi- cantly inhibited by PEE,the decreased SOD activities significantly increased with a obvious reduction of MDA.In addition,there were no obvious difference about the body weight and organ index of mice. Conclusion These results indicate PEE restore the learning and memory deficiency induced by scopolamine via inhibition of the AChE actitivies and in- creasing the antioxidant level,and it may be developed for prevention or treatment of AD.
