摘要
目的探讨腹膜透析液(PDF)对人腹膜间皮细胞(HPMCs)周期和周期蛋白p16的影响及骨髓间充质干细胞(BMSCs)的干预作用。方法从择期行腹膜透析置管手术者大网膜和PDF中分离培养出HPMCs,比较细胞形态、传代数,了解PDF对HPMCs早衰的促进作用。将PDF加入常规培养的HPMCs,通过HPMCs传代数、细胞周期、p16的检测以观察PDF对HPMCs早衰的影响及可能机制。在培养液中加入BMSCs,观察BMSCs对HPMCs周期和周期蛋白的影响。结果与对照细胞相比,PDF致HPMCs传代数减少4~5代,细胞周期阻滞于G_1期,细胞形态呈早衰细胞样变化,p16表达增多。PDF中分离培养出的HPMCs接近早衰细胞,加入BMSCs培养的HPMCs早衰进程延缓。结论 BMSCs可通过影响细胞周期和周期蛋白来延缓PDF所诱导的HPMCs的早衰进程。
Objective To investigate the effect of peritoneal dialysis fluid(PDF)on cell cycle of human peritoneal mesothelial cells(HPMCs)and cyclin p16 and the intervention of bone marrowderived mesenchymal stem cells(BMSCs)on it.Methods HPMCs were isolated from the peritoneal tissue and PDF from the patients undergoing peritoneal dialysis.Then HPMCs were cultured with RPMI-1640 medium in vitro.The effects of PDF on cellular senescence of HPMCs were preliminarily examined by observing cell morphology and cell generation.The PDF was added into the conventional cultured mesothelial cells.The effects of PDF on cellular senescence of HPMCs were examined by observing cell generation,cell cycle and p16.After BMSCs were added into the culture medium,the effects of BMSCs on the senescence of HPMCs were observed for understanding the mechanism.Results Compared with the control cells,HPMCs cultured in PDF showed decreased cell generation by four to five generations and the cell cycle was stopped at G_1 phase.It also showed that p16 expression was increased and the morphological changes of senescent cells appeared.Conclusion Through affecting cell cycle and cyclin,BMSCs may delay the process of cellular senescence of HPMCs induced by PDF.
