摘要
目的 探讨复合型结节性硬化症2(TSC2)基因的表达产物马铃薯球蛋白(Tuberin)在胃癌组织中的表达及其临床意义.方法 应用原位杂交和免疫组织化学链霉菌抗生物素蛋白-过氧化物酶(SP)法检测197例胃腺癌组织及65例正常胃黏膜组织中Tuberin mRNA和蛋白的表达,并分析其表达与胃癌临床病理特征的关系.结果 Tuberin在正常胃黏膜组织及癌组织中mRNA表达率分别为80.0%(52/65)及38.1% (75/197),组间比较差异有统计学意义(x2=34.401,P<0.05);正常黏膜组织及癌组织中Tuberin蛋白表达率分别为67.7%(44/65)、34.5%(68/197),组间比较差异有统计学意义(x2 =21.978,P<0.05);胃腺癌组织中Tuberin mRNA和蛋白表达均与癌组织的TNM分期密切相关(P<0.05).此外,Tuberin mRNA和蛋白的表达具有正相关(r=0.552,P<0.05).结论 胃腺癌组织中Tuberin mRNA和蛋白均低表达,其低表达可能与胃癌发生有关.
Objective To explore the expression of tuberin mRNA and protein in gastric carcinoma tissue and its clinical significance.Methods Immunohistochemical staining [streptavid-in-peroxidase (SP) method] and in situ hybridization were used to detect tuberin protein and mRNA expression in 197 cases of gastric carcinoma and 65 cases of normal gastric epithelia.The relationship between tuberin expression and clinicopathologic features of gastric carcinoma was analyzed.Results The positive expression rate of tuberin mRNA in normal mucosa tissue and cancer tissue was 80.0% (52/65) and 38.1% (75/197) respectively.There was statistically significant difference between two groups (x2 =34.401,P 〈 0.05).The positive expression rate of tuberin protein in normal mucosa and cancer tissue was 67.7% (44/65) and 34.5% (68/197) respectively.There was statistically significant difference between two groups (x2 =21.978,P 〈 0.05).The expression of tuberin mRNA and protein was obviously correlated with the TNM grade in gastric carcinoma tissu (P 〈 0.05).Conclusion The expression of tuberin mRNA and protein was up-regulated in gastric carcinoma tissue,which may be related to the occurrence of gastric carcinoma.
