摘要
目的对广州番禺地区人群开展ABO亚型筛查和基因分型研究,了解该地区ABO亚型的分布频率和基因分型的特征。方法随机抽取本地区参加献血和临床体检样本,采用96孔微量板法快速筛查ABO血型正定型和反定型,凡是正反定型不一致的进一步做亚型鉴定;采用抗-A_1试剂对经单克隆ABO正定型试剂确定为A型和AB型的样本进行A_2和A_2B亚型筛查,采用试管法进行血清学鉴定;采用PCR-SSP法对上述鉴定为亚型样本的进行基因分型,对特殊样本进行测序分析。结果在39 106例ABO正反定型检测中,检出2例A_3亚型、基因分型均为A102/O01,1例B_3亚型、基因分型B303/O02。在6 664例经单克隆试剂定型A型中筛检出16例A_2亚型,发生频率约为0.24%(16/6 664);基因分型4例A205和2例A201,发生频率约为0.09%(6/6 664)。在834例经单克隆试剂定型AB型中筛检出44例A_2B亚型,发生频率约为5.28%(44/834),基因分型检出15例A205/B01、4例A201/B01,发生频率约为2.28%(19/834)。结论本地区人群中发现A_2、A_2B、A_3、B_3等亚型,使用PCR-SSP方法结合血清学检测更能准确鉴定ABO亚型。
Objective To study and analyze the ABO subtype frequencies and genotypic characteristics in Panyu district of Guangzhou. Methods The samples of blood donors and clinical health examinees were randomly selected. ABOblood groups were rapid screened by 96-well micro plate method.Any inconsistent sample of forward and reverse blood typing was further identified. Using anti-A_1 reagent,A_2 and A_2B subtypesscreened from samples that had been already identified as A and AB type by monoclonal ABO reagents,a test tube method was used for serological identification. The subtype samples identified above were genotyped by PCR-SSP method. Special samples were sequenced. Results In 39 106 cases of ABO forward and reverse typing samples,2 cases of A_3 subtype were detected,with genotypes A102 / O01,1 case of B_3 with genotype B303 / O02. In 6 664 type A samples identified by monoclonal ABO reagents,16 cases of A_2 were detected at a frequency of 0. 24%( 16 /6 664). Genotypes were 4 A205 and 2A201,at a frequency of 0. 09%( 6 /6 664). In 834 type AB samples identified by monoclonal ABO reagents,44 cases of A_2 B were detected at a frequency of 5. 28%( 44 /834). Genotypes were 15 A205 / B01 and 4 were A201 / B01,at a frequency of 2. 28%( 19 /834). Conclusion A_2,A_2 B,A_3,B_3,ABO subtypes were detected in the population in the region. Using the PCR-SSP method combined with serological test,ABO subtypes were more accurately identified.
