长链非编码RNA GHET1在胃癌中的表达及其临床意义 被引量:8

摘要 目的:探讨长链非编码RNA GHET1(lnc RNA gastric carcinoma high expressed transcript 1,lnc RNA GHET1)在胃癌组织和不同分化程度人胃癌细胞中的表达情况,探索其在胃癌发生、发展过程中的作用。方法:收集35例胃癌组织及癌旁组织,培养5株分化程度不同胃癌细胞系和正常胃黏膜细胞GES-1,通过实时荧光定量PCR(quantitative real-time polymerase chain reaction,q RT-PCR)检测组织和细胞中GHET1的表达量,结合临床资料分析GHET1与胃癌临床病理特征的关系。结果 :GHET1在胃癌组织中表达增高,且与胃癌的侵袭和TNM分期相关(P<0.01);在分化程度不同的胃癌细胞中GHET1均有不同程度的表达上调(P<0.05),但GHET1表达水平与胃癌细胞的分化程度不相关(P>0.05)。结论:GHET1的在胃癌中表达上调,且与侵袭和TNM分期有关,可能成为辅助胃癌诊断和预后的潜在分子标志。 Objective To investigate the expression of long non-coding RNA GHET1 in human gastric cancer tissues and cell lines, and to explore its role in the development and progression of gastric cancer.Methods Gastric cancer tissue and normal tissue samples were collected from 35 patients, and cultured five gastric cancer cell lines with different degrees of differentiation and normal gastric mucosa cell line GES-1 in vitro. The expression level of GHET1 was examined using real-time RT-PCR(q RT-PCR) in gastric carcinoma tissue and cell lines. The statistical evaluation of q RT-PCR checking has been compared with clinical features and pathological characteristics. Results GHET1 expression was increased in gastric cancer tissue, and correlated with the invasion of gastric cancer and TNM stage(P 0.01). GHET1 expression was up-regulated in different degrees of differentiation of gastric cancer cells(P 0.05), but there was no significant correlation between the expression of GHET1 and cell lines with different degrees of differentiation(P 0.05).Conclusion Our study indicated that lnc RNA-GHET1 was up-regulated in gastric carcinoma, which was involved in invasion and TNM staging, and may become a potential molecular marker for diagnosis and prognosis of gastric cancer.
