大鼠免疫器官中CD103^+CD4^(+/-)树突状细胞的分布和形态学观察 被引量:2

摘要 目的:针对树突状细胞功能差异,探讨CD103^+CD4^+亚群在大鼠各级免疫器官中的分布与形态差异。方法:运用双重免疫组化及免疫荧光双标记法通过CD103及CD4对树突状细胞各亚群进行标记,通过光镜、荧光倒置显微镜及激光共聚焦显微镜观察树突状细胞各亚群的含量、分布和形态。结果:肠系膜淋巴结中CD103^+DC约占其内细胞总数的1.7%,胸腺次之约1.2%,脾脏最少约0.98%。不同部位各亚群DC的形态也存在一定差异,胸腺及肠系膜淋巴结DC大多数为CD4^-CD103^+DC且形态不规则有较长突起。脾脏CD4^-CD103^+DC核小而不规则,突起较细小。结论:大鼠各级免疫器官中各树突状细胞亚群存在分布及形体差异,这一差异可能与其在迁移分化过程中在各级免疫发挥不同调节功能有关。 Objective: For the functional differences in dendritic cell subsets,the study gains insight into the distributional and morphological differences of rat dendritic cells in immune organs. Methods: Immunohistochemical staining and Immunofluorescent staining were perfomed to study the morphology and the distribution of rat dendritic cells by DC marker( including CD103 and CD4). Results: In Mesenteric lymph nodes CD103+DC accounted for about 1. 7% of its total number of mononuclear cells,followed by about1. 2% in the thymus and least in spleen about 0. 98%. There were certain differences among DC in different immune organs. The majority of TDCs were CD4-CD103+DC,the morphology of CD4-CD103+T DCs and MLN DCs was irregular and long dendrites could be observed. In MLN most of CD4-CD103+DCs contain a large,round nucleus and with relatively long dendrites. The nuclei of CD4-CD103+DCs were small and irregular while their dendrites was smaller. Conclusion: That the distribution and morphology of the subsets of dendritic cells in different immune organs being not identical may be related to the subsets of dendritic cells playing different regulatory functions in the process of migration and differentiation in immune organs.
