摘要
目的:探讨宫颈癌患者血清和组织中谷胱甘肽过氧化物酶(Glutathione peroxidase,GPx)活性和宫颈组织中硒结合蛋白1(Selenium Binding Protein 1,SBP1,SELENBP1)表达之间的相互关系,为SELENBP1在宫颈癌发生发展中的作用机制提供理论基础。方法:收集临床住院病人血清样本检测GPx活性,收集相应宫颈组织样本检测GPx活性及采用Western blot方法检测组织内SELENBP1蛋白表达水平,用Spearman等级相关来分析其相关性。结果:宫颈癌病人血清中GPx活性明显降低;与正常宫颈、CINⅢ级组织和相应癌旁组织相比,癌组织中GPx活性增强,SELENBP1蛋白表达也降低。结论:宫颈癌组织中SELENBP1表达水平与肿瘤组织微环境中GPx活性呈负相关,而与病人血清中GPx活性无关,提示SELENBP1可能是通过肿瘤微环境中GPx来发挥抗肿瘤作用。
Objective: To investigate the correlation between the expression of Selenium Binding Protein 1( SBP1,SELENBP1)and Glutathione peroxidase( GPx) activity in cervical cancer tissues and provide theoretical basis in the development of cervical cancer for SELENBP1. Methods: The expression of SELENBP1 was detected by Western blot in normal cervical tissues,intraepithelial neoplasia( CIN) Ⅲ,cervical cancer( CA) and related adjacent cancer( p CA) tissues,and GPx activity in serum and tissues using a spectrophotometric assay according to the instructions. Results: The GPx activity in serum of patients with cervical cancer was significantly decreased; compared with normal cervix,CINⅢ and corresponding adjacent tissues groups,GPX activity was increased,and the expression of SELENBP1 was decreased in cancer tissues. Conclusion: The role of SELENBP1 in cervical cancer development and prognosis,which can play an anticancer role through GPx in tumor microenvironment.
