地尔硫抑制小鼠吗啡戒断反应及血浆和脑匀浆一氧化氮的含量 被引量:2

摘要 目的··:观察钙拮抗剂盐酸地尔硫对小鼠吗啡戒断反应及血浆和脑匀浆一氧化氮 (NO)含量的影响。方法··:皮下注射定量吗啡 ,建立小鼠吗啡依赖动物模型 ;腹腔注射纳洛酮催促 ,根据小鼠戒断反应中出现跳跃反应的潜伏期、跳跃次数和体重丢失评定戒断反应的强度 ;用硝酸还原法检测其血浆和脑内NO含量。结果·· :3种不同剂量 (25、50、100mg·kg-1)的盐酸地尔硫对小鼠吗啡戒断反应有明显的抑制作用 ,并呈量效关系;100mg·kg-1的盐酸地尔硫可抑制吗啡戒断反应引起的血浆和脑匀浆NO含量的升高。结论·· :盐酸地尔硫可抑制小鼠吗啡戒断反应 ,并能抑制戒断反应引起的血浆和脑匀浆NO含量的升高。 Objective:To observe the effect of diltiazen hydrochloride on morphine withdrawal syndromes and the level of NO in plasma and brain homogenate in morphine dependent mice. Methods :A physical dependent model in mice was established by subcutaneous injection of morphine. After intraperitoneal injection of noloxone, the intensity of withdrawal syndromes was evaluated according to the jumping latency, jumping number and the loss of weight. The level of NO was detected with Griess method. Results:The physical withdrawal syndromes in morphine dependent mice were inhibited partly by three different doses (25, 50, 100 mg·kg-1)of diltiazem and showed a significant dose-dependent manner. The increased level of NO in plasma and brain homogenate in morphine dependent mice could be reduced by large dose (100 mg·kg-1)of diltiazem. Conclusion: The withdrawal syndromes and the level of NO in plasma and brain homogenate in morphine dependent mice could be inhibited obviously by diltiazem.
