摘要 目的评价火炬树叶乙醇提取物(TLEEP)的降血糖活性。方法体外实验,检测TLEEP对α-葡萄糖苷酶活性的抑制。体内实验,雄性昆明小鼠除空白组外,其余采用链脲佐菌素诱导小鼠糖尿病模型,分为阳性对照组1(阿卡波糖组,6.66μg·g^(-1)·d^(-1)),阳性对照组2(消渴丸组,1 390μg·g^(-1)·d^(-1)),TLEEP低、中、高剂量组(200、400、800μg·g^(-1)·d^(-1))。每组10只,连续干预3周。给药结束后眼眶取血检测血清胰岛素、血脂(TG、TC、HDL-C、LDL-C)、肝糖原含量,并检测小鼠的糖耐量。结果体外实验结果表明,TLEEP在0.4 g·L^(-1)时对α-葡萄糖苷酶活性的抑制率达到91%,具有明显的α-葡萄糖苷酶抑制作用。体内实验结果表明,TLEEP低、中、高剂量组均具有降低高血糖小鼠血糖的作用,其中以TLEEP高剂量组(800μg·g^(-1)·d^(-1))的降血糖作用最为明显,同给药前相比下降55%,TLEEP能够显著升高高血糖小鼠的胰岛素和肝糖原含量,降低小鼠血清中TC、TG、LDL-C含量(P<0.05),明显提高HDL-C含量(P<0.05)。结论 TLEEP可显著降低STZ致糖尿病小鼠血糖,改善其脂代谢紊乱症状,是一种具有较大潜力的降血糖的天然药物。 Objective To estimate the hypoglycemic effects of the alcohol extract from the leaves of Rhus Typhina( TLEEP) in vitro and in vivo. Methods In vitro experiment,the effects of TLEEP on the activity of alpha-glucosidase were observed. In vivo experiment,diabetic model was induced by streptozocin( STZ)in male Kunming mice. The model rats were then divided into the following groups: model control group,positive control group 1( acarbose group 6. 66 μg·g(-1)·d(-1)),positive control group 2( Xiaoke pill group 390 μg·g(-1)·d(-1)) and low-,medium-,high-dose of TLEEP group( 200,400,800 μg·g(-1)·d(-1)).Each group had 10 rats,and received 3-week treatment. After treatment,the blood was taken from the eyes to detect serum insulin,blood lipid( TG,TC,HDLC,LDL-C),hepatic glycogen,and the sugar tolerance. Results The results of in vitro experiment showed that TLEEP( 0. 4 g·L(-1)) inhibited the activity of alpha-glucosidase by 91%,with significant inhibition of alpha-glucosidase activity. The results of in vivo experiments showed that TLEEP,especially the high-dose TLEEP significantly reduced the blood glucose of model mice. The TLEEP significantly improved the serum insulin level and hepatic glycogen content,reduced serum levels of TC,TG and LD-C( P 〈 0. 05),and improved the HDL-C content( P 〈 0. 05) of model mice. Conclusion TLEEP can markedly decrease blood glucose and improve the lipid metabolism disorders in STZ-induced diabetic mice. The TLEEP is a natural herb with potential hypoglycemic activities.
