摘要
目的分析乳腺癌组织中Eph A2、SIAH2、β-catenin的表达量及其与肿瘤恶性程度的相关性。方法将2012年8月至2015年12月间接受乳腺肿瘤切除治疗的患者128例作为研究对象,根据肿瘤的良恶性不同分为乳腺癌组54例,乳腺腺瘤组74例。将手术切除的肿瘤组织碾磨成匀浆,并应用Western-blot蛋白表达检测及酶联免疫吸附法(ELISA)血清学检测,对比两组患者的肿瘤组织Eph A2、SIAH2、β-catenin表达,肿瘤组织增殖相关指标,血清侵袭转移相关指标等水平差异,并进一步分析肿瘤组织Eph A2、SIAH2、β-catenin表达与恶性程度的相关性。结果乳腺癌组患者的肿瘤组织Eph A2、SIAH2、β-catenin表达量均显著高于乳腺腺瘤组患者(P<0.05);乳腺癌组患者的肿瘤黑素瘤抗原(MAGE)-A11、FOXM1、细胞增殖抗原标记物(Ki67)、细胞周期素D1(Cyclin D1)表达量高于乳腺腺瘤组患者,E-cadherin表达量低于乳腺腺瘤组患者(P<0.05);乳腺癌组患者的血清肝细胞生长因子(HGF)、转化生长因子-β(TGF-β)、Six1、碱性成纤维细胞生长因子(b FGF)、胸苷激酶1(TK1)、中期因子(MK)值均显著高于乳腺腺瘤组患者(P<0.05);乳腺癌组织的Eph A2、SIAH2、β-catenin表达量与MAGE-A11、FOXM1、Ki67、Cyclin D1、HGF、TGF-β、Six1、b FGF、TK1、MK值呈正比,与E-cadherin值呈反比(P<0.05)。结论乳腺癌组织中高表达的Eph A2、SIAH2、β-catenin与肿瘤的增殖及侵袭转移等特性均关系密切,可以作为宏观判断肿瘤恶性程度的有效指标。
Objective To analyze the expression of EphA2,SIAH2 and β- catenin in breast cancer and its correlation with the degree of malignancy. Methods 128 patients received breast tumor resection in our hospital from August 2012 to December 2015. All these patients were selected as the research object. According to the difference of benign and malignant tumors,patients were divided into breast cancer group( 54 cases) and breast adenoma group( 74 cases). Resected tumor tissue were milled into homogenate. The Western blot protein expression detection and enzyme- linked immunosorbent assay( ELISA) serological detection were used. The difference of EphA2,SIAH2,β- catenin expression in tumor tissue,proliferation related indexes in tumor tissue,serum invasion related indicators between the two groups were compared and the correlation between EphA2,SIAH2,β- catenin expression in tumor tissue and degree of malignancy were further analyzed. Results EphA2,SIAH2,β- catenin expression in tumor tissue of breast cancer groups patients were significantly higher than those of breast adenoma group patients( P〈0. 05). MAGE- A11,FOXM1,Ki67,cyclin D1 expression in tumor tissue were higher than breast adenoma groups. E- cadherin expression was lower than adenoma group( P〈0. 05). Serum HGF and TGF- β,Six1,bFGF,TK1,MK values of breast cancer groups were significantly higher than those of the breast adenoma group( P〈0. 05). EphA2,SIAH2,β- catenin expression in breast cancer tissue were positively correlated with MAGE- A11,FOXM1,Ki67,cyclin D1,HGF and TGF- β,Six1,bFGF,TK1,MK values,negatively correlated with E- cadherin( P〈0. 05). Conclusion High expression of EphA2,SIAH2 and β- catenin in breast cancer is closely related to the proliferation and invasion and metastasis of tumor. It can be used as an effective indicator to judge the malignant degree of tumor.
