环孢素A抑制大鼠腹主动脉球囊损伤后内膜增生

Inhibitions of Cyclosporin-A on Intima Proliferation of Rat Abdominal Aortas Injured by Balloon

摘要 观察环孢素A防止血管内膜增生作用与单核细胞趋化蛋白-1(Monocyte chemoattractant protein-1,MCP-1)水平的关系。雄性SD大鼠36只,随机分为假手术组(n=12)、手术组(n=12)、干预组(n=12)。术后30 d取实验材料,血管组织作HE染色;荧光定量PCR技术检测损伤血管壁钙调神经磷酸酶(Calcineurin,Ca N)、活化T细胞核因子3(Nuclear factor of activated T cell,NFATc3)、MCP-1在mRNA水平上的表达变化;ELISA法测定血浆MCP-1在蛋白水平上的表达变化。假手术组内膜无增生,手术组血管壁内膜明显增生,干预组内膜亦出现增生,但内膜增生、内膜/中膜厚度与手术组相比较明显减轻(P<0.05)。干预组与手术组相比较,血管壁组织Ca N、NFATc3、MCP-1 mRNA表达明显减少(P<0.05)。血浆MCP-1水平干预组与手术组相比明显降低(P<0.05)。环孢素A通过抑制Ca N-NFATc信号通路减轻受损血管壁以及血浆的炎症因子MCP-1水平,从而减轻球囊损伤术后动脉内膜增生。 To observe the effect of cyclosporin-A on intima proliferation,the arterial wall and serum monocyte chemoattractant protein-1( MCP-1) levels of rat abdominal aortas injured by balloon. Thirty-six adult male SD rats were randomly divided into three groups: sham group( n = 12),operation group( n = 12) and intervention group( n = 12). Aortic tissue was harvested at day 30 after balloon injury for pathological with HE staining. The mRNA expressions of Calcineurin( Ca N),Nuclear factor of activated T cell 3( NFATc3),MCP-1 in the aortic tissue were measured by Real time PCR. The levels of MCP-1 were detected by Elisa. Aortic tissue showed intimal thickening in operation group,while the abdominal aortas in the sham group were not. Compared with the operation group,the intimal thickness,the ratio of the intimal and the medial layers was decreased significantly( P 0. 05) in the intervention group. Rea1 time PCR showed that the expression of Ca N,NFATc3,MCP-1 mRNA in intervention group was decreased significantly compared with operation group( P 0. 05). The serum concentrations of MCP-1 of intervention group are lower than those of operation group( P 0. 05). Cyclosporin-A can inhibit the intima proliferation by supressing Ca N-NFATc signal transduction pathway,and decreasing the arterial wall and serum inflammatory cytokines MCP-1 levels in aorta-injured rats.
