广州与佛山地区2014年登革病毒基因型特征研究 被引量:4

Genotypic characteristics of dengue virus in Foshan and Guangzhou in 2014

摘要 目的对2014年广州与佛山地区登革病毒C-prM区进行基因测序,从分子水平与相关国际流行株进行同源性比较,构建系统发育树,追踪其可能输入来源。方法收集本院2014年9月-11月447例登革热疑似患者急性期血清,随机选取其中广州与佛山地区不同区域的24例标本,提取病毒RNA,RT-PCR法扩增C-prM基因,并将其克隆到p MD19T载体,测序并利用生物信息学软件进行分析。结果成功测序登革病毒11株,BLAST分析表明均为登革1型病毒(DENV-1)。系统发育树结果显示GZ2014-01等序列与2010年印度德里流行株同源性高(相似度为99.21%),GZ2014-02等序列与2013年广州流行株相近(相似度为97.64%~98.82%)。广州株GZ2014-01等和佛山株GZ2014-23属于基因型Ⅴ,而广州株GZ2014-02等和佛山株GZ2014-20属于基因型Ⅰ。结论 GZ2014-01等株可能来源于印度德里,而GZ2014-02等株可能来源于中国广州。登革病毒多基因型流行可能是造成2014年登革热大暴发的原因之一。 Objective To conduct gene sequence on the C- prM zone of dengue virus in Foshan and Guangzhou in 2014 for homology comparison on molecular level and the epidemic strains,so as to draw the phylogenetic tree to trace their potential origins. Methods The serums were collected from acute patients who were reported for clinically suspected dengue infection in our hospital during September- November 2014,and viral RNAs were extracted from 24 cases randomly selected from Foshan and Guangdong. C- prM gene was amplified by RT- PCR,and cloned into p MD19 T vector,and then sequenced for analysis with molecular phylogenetic software. Results 11 samples were sequenced successfully,and confirmed as DENV- 1 by BLAST. The GZ2014- 01 sequence had highly similarity on homology with the the epidemic strains from Delhi of Indian in2010( similarity of 99. 21%). While GZ2014- 02 sequences were similar with the epidemic one in 2013,Guangzhou( similarity among 97. 64%- 98. 82%). Guangzhou strain GZ2014- 01 and Foshan strains GZ2014- 23 belong to genotype V,and Guangzhou strain GZ2014- 02 and Foshan strains GZ2014- 20 belong to genotype I. Conclusion GZ2014- 01 and other strains may be derived from Delhi of India,and GZ2014- 02 and other strains may be derived from Guangzhou. The multi- gene epidemic of Dengue virus genotype may be one of the causes of the dengue fever outbreak in 2014.
