摘要
目的检测鸡胚细胞中微生物污染情况,初步分析污染源头并提出相应的防污染措施。方法利用API系统分别对分离自原代鸡胚成纤维细胞中的3种常见的微生物菌株进行鉴定。结果经鉴定3株污染菌株分别为木糖氧化产碱菌(Alcaligenes xylosoxidans)、蜡样芽胞杆菌(Bacillus cereus)和金黄色葡萄球菌(Staphylococcus aureus)。结论从鉴定的结果分析,产生微生物污染的因素是多方面的,但其中工作人员没有严格执行操作规范,自身携带的微生物是导致微生物污染的主要因素,其次是洁净室环境、设施及日常清洁消毒工作不到位。因此,严格执行各项规章制度,规范人员操作,彻底消毒洁净室通风系统和相关设施并做好日常清洁消毒工作对于防止原代细胞污染至关重要。
Objective The microbial contamination in primary chicken embryo fibroblast cells was investigated to analyze the pollution source and formulate the corresponding control measures. Methods Three common microbial strains from primary chicken embryo fibroblast cells were isolated and identified by API system. Results The three microbial strains were Alcaligenes xylosoxidans,Bacillus cereus and Staphylococcus aureus,respectively. Conclusion Several factors could account for microbial contamination in cultured cells according to the test results. The staff who did not strictly follow the operation standards were the main contamination factor. Furthermore,the clean room environment,facilities and daily cleaning and disinfection work were unqualified. Therefore,the strict implementation of the rules and regulations,standardization of personnel operation,thorough cleaning and disinfection of clean room ventilation system and related facilities and daily cleaning and disinfection work are essential to control primary cell cultures pollution.
