同源异型盒基因HoxD10对鼻咽癌恶性生物学行为的调控作用研究 被引量:2

摘要 目的通过检测HoxD10基因在鼻咽癌细胞及正常鼻咽上皮细胞中的表达水平,及其对鼻咽癌细胞增殖、侵袭力及皮下移植瘤的影响,探讨其在鼻咽癌的发生、发展过程中的作用。方法利用Western blot检测HoxD10基因在永生化非癌性人鼻咽上皮细胞NP-69,鼻咽癌细胞株CNE-1及过表达HoxD10基因的5-8F细胞中表达情况;利用CCK-8、Transwell及皮下移植瘤构建鼻咽癌裸鼠模型,分别检测5-8F细胞过表达HoxD10基因后增殖、侵袭力的变化情况及对裸鼠移植瘤生长的影响。结果 2组鼻咽癌细胞的HoxD10蛋白表达水平显著低于NP-69细胞,差异有统计学意义(P<0.01);过表达HoxD10基因后5-8F细胞的增殖、侵袭力显著降低,并能显著降低裸鼠移植瘤的瘤体体积和重量,差异均有统计学意义(P<0.01)。结论 HoxD10基因在鼻咽癌细胞中低表达,发挥着抑癌基因作用,HoxD10基因过表达可以显著降低鼻咽癌细胞的增殖侵袭能力,HoxD10基因的表达缺失可能参与了鼻咽癌的发生发展及侵袭转移过程,有望成为诊断和治疗鼻咽癌的一种新的标记物。 Objective To explore the role of HoxD10 expression in nasopharyngeal carcinoma cells and normal nasopharyngeal epithelial cells on the regulation of human nasopharyngeal carcinoma proliferation and invasion,and to investigate the role of it in the occurrence and development of nasopharyngeal carcinoma. Methods We used Western blot to detect the expression of HoxD10 in NP- 69,CNE- 1,5- 8F and the overexpression of HoxD10 in 5- 8F cell. The ability of 5- 8F cell proliferation was studied by CCK- 8 and invasion by Transwell. To establish subcutaneous transplanted model of human nasopharyngeal carcinoma in nude mice and to explore the effect of HoxD10 on the growth of transplanted tumor. Results The protein expression levels of HoxD10 was significantly lower than that in the NP- 69 cells in the 2 groups of nasopharyngeal carcinoma cells,with the difference statistically significant( P 〈0. 01). After HoxD10 gene overexpression,the proliferation and invasion force of HoxD10 and 5- 8F cells decreased significantly,and can significantly reduce the volume and weight of the transplanted tumor in nude mice,with the difference statistically significant( P 〈0. 01). Conclusion Our study suggested that HoxD10 is lower expression and act as a tumor- suppressor gene in asopharyngeal carcinoma. Overexpression of HoxD10 inhibits proliferation and invasion in 5- 8F cell line. The low expression of HoxD10 is significantly related with the occurrence and progress of nasopharyngeal carcinoma,which may be a new target of gene therapy of asopharyngeal carcinoma.
