Reprimo基因G824C和C839G多态性与肺癌易感性的关系

摘要 目的探讨Reprimo基因3'非翻译区G824C和C839G多态性与肺癌发生的关系。方法等位基因特异性PCR(AS-PCR)检测36例肺癌患者(肺癌组)和其癌旁正常组织(对照组)的Reprimo基因3'非翻译区G824C位点和C839G位点基因多态性,分析其与肺癌易感性之间的关系。结果经Hardy-Weinberg平衡检验显示,在G824C位点对照组和癌症组的基因型(GG,CG和CC)观测(理论)频数分布均不符合Hardy-Weinberg平衡(P=0.042和0.003);在C839G位点对照组和癌症组的基因型(CC,CG和GG)观测(理论)频数分布均不符合Hardy-Weinberg平衡(P=0.000和0.000)。与GG型相比,带有C等位基因个体(CG型和CC型)在G824C位点增加个体患肺癌的发病风险不明显(OR=1.218,95%CI:0.572-2.594);与CC型相比,GG型在C839G位点可能会增加个体患肺癌的发病风险(OR=4.000,95%CI:0.733-21.830)。结论携带Reprimo基因3'非翻译区C839G位点GG基因的个体患肺癌的发病风险可能增加。 Objective To investigate the association of Reprimo 3'-untranslated region(3'-UTR)G824C and C839 Gpolymorphisms with susceptibility to lung cancer. Methods Allele specific- PCR(AS- PCR)was used to detect thepolymorphisms in the 3'- UTR sites G824 C and C839 G of Reprimo gene in the lung tissue samples from 36 lung cancerpatients and corresponding paracancer tissues. Statistical analysis was applied to demonstrate the association between thegenotype frequencies and lung cancer susceptibility. Results Hardy-Weinberg equilibrium test indicated that they were outwith the equilibrium in the observation frequencies and theoretical frequencies of G824 C genotypes(GG, CG and CC)in thecontrol and lung cancer tissues(P=0.042 and 0.003, respectively). Similar results were also obtained in the site C839 G whencomparing genotypes(CC, CG, GG)in control group and lung cancer group(P=0.000 and 0.000, respectively). Compared withthe patients with GG genotype, the individuals carrying allele of C(namely CG genotype and CC genotype)had no significantgenetic susceptibility to lung carcinoma(OR=1.218, 95% CI: 0.572-2.594). However, the subjects with the GG genotype mighthave an increased risk of lung cancer compared with those of CC genotype in site C839G(OR=4.000, 95% CI: 0.733-21.830).Conclusion The individuals carrying GG genotype at the site C839 G in the 3'-UTR of Reprimo might have higher geneticsusceptibility to lung cancer.
