益智仁醚提取物对糖尿病肾病小鼠氧化应激的影响 被引量:9

Effect of Alpiniae Oxyphyllae Fractus ether extract on oxidative stress in mice with diabetic nephropathy

摘要 目的研究益智仁醚提取物对糖尿病肾病小鼠的抗氧化作用。方法以糖尿病肾病小鼠为研究对象,通过制模分组,设为正常对照组、模型组、厄贝沙坦组、益智仁高、中、低剂量组,厄贝沙坦组以10 mg/(kg.d),益智仁高、中、低剂量组分别以20 g/(kg·d)、10 g/(kg.d)、5 g/(kg.d)给小鼠灌胃。正常对照组、模型组以等体积蒸馏水灌胃。实验至第4周末,检测不同组小鼠的血清SOD、MDA、CAT、GSH-PX,比较各组之间差异。结果益智仁醚提取物高、中剂量组可以升高血清中SOD、CAT、GSH-PX活力,降低MDA含量,与模型组比较差异有统计学意义(P<0.05)。与厄贝沙坦组比较差异无统计学意义(P>0.05)。而低剂量组中各个血清氧化应激指标变化与模型组差异无统计学意义(P>0.05)。结论益智仁醚提取物高、中剂量组能够改善糖尿病肾病小鼠的氧化应激指标,具有一定的抗氧化作用,与已证实有抑制氧化应激作用的厄贝沙坦相仿。 Objective To determine the antioxidant effect of Alpiniae Oxyphyllae Fractus ether extract on the mousemodel of diabetic nephropathy. Methods Mouse model of diabetic nephropathy was established by intraperitoneal injectionof streptozotocin(STZ)followed by high-fat diet,and identified by blood glucose level and proteinuria. The mice were dividedinto normal control group,model group,irbesartan group [10 mg/(kg·d)],and high-,medium- and low-dose of the etherextract groups [20,10 and 5 g/(kg·d)]. The mice of the irbesartan group and the treatment groups were given correspondingagent respectively by gavage,while those of normal control and model group were treated with equal volume of distilled water.After 4 weeks' treatment,the serum levels of superoxide dismutase(SOD),malondialdehyde(MDA),catalase(CAT)andglutathione peroxidase(GSH-PX)were measured and compared in different groups. Results The medium- and high-dosesof the ether extract increased the activities of SOD,CAT and GSH-PX in the serum,while decreased the content of MDA,withsignificant differences from the model group,but there was no significant differences with irbesartan group. No significantdifferences were found in the above indexes between the model group and low-dose group. Conclusion Medium- and high-doses of Alpiniae Oxyphyllae Fractus ether extract improve the oxidative stress indexes in diabetic nephropathy mice,and exertcertain antioxidant effect. The effect is similar to that of irbesartan,which has been proved to have the inhibitory effect onoxidative stress.
