侵染甜椒的番茄黄化曲叶病毒的分子鉴定和全序列分析 被引量:2

Molecular identification and full-length sequence analysis of Tomato yellow leaf curl virus on sweet pepper in China

摘要 番茄黄化曲叶病毒(Tomato yellow leaf curl virus,TYLCV)在世界范围内可危害多种作物,造成植株矮化、叶片皱缩变形、局部黄化等症状。该病毒自1964年首次报道以来已蔓延至世界多地。在我国2006年上海首次报道该病毒,随后江苏、山东、安徽、北京、河北、天津等地相继报道,危害严重。 Tomato yellow leaf curl virus(TYLCV),a single-stranded DNA virus transmitted by whiteflies,causes severe damage to tomato and pepper production worldwide.During the surveying of viral disease on vegetables in 2014,yellowing and slightly curl symptom on upper leaves was observed in sweet pepper plants growing in a greenhouse of Haidian district,Beijing,China.To test whether these diseased sweet pepper plants were infected by geminiviruses,a PCR assay was performed with total DNA isolated from diseased sample using a pair of degenerated primers PA/PB for geminiviruses.A fragment(about 540 bp) was amplified and sequence analysis indicated that the sample might be infected by TYLCV.The isolate was named TYLCV-Haidian.Complete genome sequence of TYLCV-Haidian isolate was obtained,which is 2 781 nt in length.Phylogenetic analysis showed that TYLCV-Haidian clustered with TYLCV-SH2,an isolate in Shanghai.To our knowledge,this is the first report of complete sequence of TYLCV in sweet pepper of Beijing,China.
