润生方对人胃癌细胞SGC-7901生物学活性的影响及诱导凋亡研究

Run sheng Recipe on biological characters of gastric carcinoma SGC-7901 Cells and the research about induce apoptosis of it

目的观察润生方体外作用于人胃癌细胞株SGC-7901后对其增殖、迁徙、侵袭以及诱导凋亡能力的影响。方法采用MTT法、划痕实验、Transwell小室法、Annexin-V/PI双染法及RT-PCR法检测润生方对SGC-7901细胞增值、迁徙、侵袭及诱导凋亡能力的影响。结果与对照组比较,润生方有抑制人胃癌细胞SGC-7901增殖的作用(P<0.01),且呈现时间和浓度依赖性。润生方作用人胃癌细胞SGC-7901后,侵袭的细胞数较对照组明显减少(P<0.01)。润生方能诱导SGC-7901凋亡,下调抗凋亡基因Bcl-2、Bcl-x L、上调促凋亡相关基因Bax、Bak。结论润生方对人胃癌细胞SGC-7901的增殖具有抑制作用;能抑制人胃癌细胞SGC-7901的迁徙和侵袭能力,并能通过下调抗凋亡基因Bcl-2、Bcl-x L、上调促凋亡相关基因Bax、Bak而诱导胃癌细胞株SGC-7901凋亡。

Objective To observe the Run sheng Recipe on Inducing Cells＇ Proliferation,invasion,migration capabilities and Cell Apoptosis of Human Gastric Carcinoma Cell Line SGC-7901. Methods Using MTT,transwell room,wooding healing,to observe the effect after treated with Run sheng Recipe including proliferation,invasion and migration capabilities,and evaluate the effect of carcinoma cell apoptosis were detected by Annexin V / PI. Results Compared with the negative control group,the treated with Run sheng Recipe group inhibited the proliferation of SGC-7901 cells,which increased with the Concentration of Run sheng Recipe and response time increasing（ P〈0. 01）.The Run sheng Recipe decreased the migration capabilities and reduce the invasion of SGC-7901 cells（ P〈0. 01） by the wooding healing and Transwell Chambers method. Treated with Run sheng Recipe,the SGC-7901 cell apoptosis was increased obviously by promoting Bak and Bax expression,inhibiting Bcl-x L and Bcl-2 expression. Conclusion Run sheng Recipe could inhibit the proliferation,decreased the migration,reduce the invasion and induces apoptosis of SGC-7901 cells.
