摘要
目的建立防风多糖提取、分离、纯化及其含量测定方法。方法以水提醇沉法制取防风粗多糖,以三氟三氯乙烷法除蛋白,用D280阴离子交换树脂柱层析脱色,采用紫外分光光度法测定防风总多糖含量。结果紫外光谱全波长扫描结果显示,防风多糖与葡萄糖标准品加入显色剂后最大波长均为486 nm,且在260和280 nm处没有吸收峰。防风多糖平均回收率为98.54%,RSD为1.09%。防风中多糖含量为2.98%。结论采用本实验提取方法可有效提取分离出纯化的防风多糖,利用紫外分光光度法测定防风结果准确,可作为防风总多糖含量测定的方法。
Objective To establish a wind polysaccharide extraction,separation,purification and its content determination method. Methods Water extract- alcohol precipitation in the legal system in wind crude polysaccharide,with three fluorine trichloroethane method in addition to the protein,with D280 anion exchange resin column chromatography decoloring,using ultraviolet spectrophotometry windproof total polysaccharide content. Results UV spectral full wavelength scanning,according to the results of wind polysaccharide and glucose standard adding chromogenic agent after the maximum wavelength of 486 nm,and no NNL in 260 and 260 nm absorption peak. Wind polysaccharide the average recovery was 98. 54%,RSD was 1. 09%. Wind in the polysaccharide content was 2. 98%. Conclusion Using the method of this experiment to extract,separate the wind of the purified polysaccharide can be extracted effectively by using ultraviolet spectrophotometry wind results are accurate and can be used as a method for the determination of total polysaccharide content against wind.
