摘要 目的探讨调控Wnt5a/Ca^(2+)/Calcineurin/NFAT通路对黑素瘤细胞增殖、侵袭能力的影响。方法体外培养慢病毒转染的黑素瘤稳转细胞系;Western blot法检测稳转细胞系中Wnt5a,NFAT,Calcineurin和VEGF蛋白的表达;CCK-8法检测稳转细胞系的增殖能力;Transwell小室实验检测稳转细胞系的侵袭能力。结果 Wnt5a蛋白的表达与Calcineurin,NFAT,VEGF蛋白的表达具有正相关性;上调Wnt5a的表达可使黑素瘤细胞增殖、侵袭能力增强,而下调Wnt5a的表达则可降低黑素瘤细胞的增殖和侵袭能力。结论调控Wnt5a/Ca^(2+)/Calcineurin/NFAT通路可影响黑素瘤细胞的增殖与侵袭,该信号通路可能参与了黑素瘤的发生、发展过程。 Objective To investigate the role of Wnt5afCa2+/Calcineurin/NFAT pathway in the proliferation and inva- sion of melanoma cells. Methods Melanoma cell line was cultured and transfected with lentivirus expressed WntSa. Western blot was used to analyze the expression of Wnt5a, NFAT, Calcineurin and VEGF. The pro- liferation of melanoma cells was determined by CCK-8 assay. Transwell cell invasion assay was used to ana- lyze the invasion of melanoma cells. Results Our results showed that the expression of WntSa was positive- ly correlated with the expression of NFAT, Calcineurinand and VEGF expression in transfected melanoma cells. Up-regulation of Wnt5a enhanced the proliferation and invasion in melanoma cells, while down-regula- tion of WntSa suppressed the proliferation and invasion of melanoma cells. Conclusion Proliferation and in- vasion of melanoma cells can be affected by the regulation of WntSa/Ca2+/Caleineurin/NFAT pathway, and this signal pathway may be involved in the progression of melanoma.
