摘要 磷脂氢谷胱甘肽过氧化物酶(PHGPx)具有明显的抗氧化活性,避免细胞膜受到过氧化损伤。本研究利用RACE技术,克隆出山参phgpx基因全长c DNA。序列分析表明:该克隆片段全长490 bp,编码162个氨基酸,有3个真核生物PHGPx特有的保守亚结构域。该片段编码的蛋白为跨膜亲水性稳定蛋白质,与柑橘、蓖麻等PHGPx氨基酸序列同源性分别为76%和75%。同时建立了9种植物的系统进化树。 Phospholipid hydroperoxide glutathione peroxidase(PHGPx) has significant antioxidant activity, avoiding the harm by peroxidation in the cell membrane.To clone the full-length cDNA encoding PHGPx gene from Panax ginseng,the RACE technology was used in this study.The sequence analysis showed that: the full length of the cloned fragment was 490 bp.lt encoded 162 amino acids and contained 3 sub-conservative domains which were specific in eukaryonic phospholipid hydroperodixe glutathione peroxidase.The encoding protein was a stable non- transmembrane hudrophilic protein.The sequence of amino acids was similar with PHGPx of citrus and castor, homologous rates of amino acids were 76% and 75%. And then, the phylogenetic tree of nine plants was constructed.
