摘要
【目的】探讨长链非编码RNA MDC1-AS在宫颈癌及癌旁组织中的表达差异及其与临床病理特征关系,并进一步研究其在宫颈癌细胞中的功能。【方法】应用q RT-PCR方法检测108例宫颈癌组织及相应癌旁组织长链非编码RNAMDC1-AS的表达,分析其与临床病理特征的关系。转染MDC1-AS表达质粒,构建过表达MDC1-AS的稳定宫颈癌Si Ha、He La细胞株,应用CCK8检测细胞增殖能力变化;Transwell实验检测过表达MDC1-AS对宫颈癌细胞侵袭、迁移能力的影响。【结果】108例宫颈癌患者中,长链非编码RNAMDC1-AS在72例癌组织的相对表达量低于癌旁组织,其低表达与淋巴结转移和淋巴脉管间隙浸润有关(P<0.05);成功建立稳定过表达MDC1-AS的Si Ha和Hela细胞株,其细胞增殖均比阴性对照显著降低(Si Ha:48 h,P<0.01;72 h,P<0.001;Hela:48 h,72 h,P均<0.001);与阴性对照相比,过表达MDC1-AS的Si Ha及He La细胞在Transwell迁移(46.3±3.0 vs 149.7±3.1;27.5±4.2 vs 85.0±3.2)、侵袭实验(55.1±0.6 vs 112.4±2.9;66.1±1.9 vs172.0±4.3)中穿膜细胞均明显减少(P均<0.01)。【结论】宫颈癌组织中长链非编码RNAMDC1-AS呈低表达,与淋巴结转移和淋巴脉管间隙浸润有关,并可能参与了宫颈癌细胞的增殖、侵袭和迁移。
[ Objective ] The aim of this study was to investigate the expression, the clinicopathological significance, and the biological role of long non-coding RNA MDC1-AS (lncRNA MDC1-AS) in the cervical cancer ceils. [Methods] Different expressions of lncRNA MDC1-AS between cervical cancer tissues and their corresponding adjacent normal tissues in 108 patients was analyzed by real-time PCR. The correlation between the expression of lncRNA MDC1-AS and their clinicopathological features was analyzed by statistical analysis. MDC1-AS plasmid was used to up-regulate the MDC1-AS expression in cervical cancer cells (Hela and Siha). The effects of MDC1-AS on the migration and invasion ability of cervical cancer cells was measured by Transwell assay. [Results] The expression of lncRNA MDC1-AS was down-regulated in tumor tissues than that in the para-cancer tissues in 72 out of 10g patients. The lower expression of lncRNA MDCI-AS was significantly correlated with lymphatic metastasis and lympho-vascular space invasion (P 〈 0.05). Moreover, after being transfected with MDC1-AS plasmid, SiHa and HeLa cells with high expression of MDC1-AS showed decreased proliferation (P 〈 0.01 )and lower ability of migration and invasion (P 〈 0.05). [ Conclusion ] Expression of lncRNA MDC1-AS was down-regulated in cervical cancer tissues, which is correlated with lymphatic metastasis and lympho-vascular space invasion. Decreased expression of lncRNA MDC1-AS might play an important role in migration and invasion ability of cervical cancer ceils.
