摘要
目的探讨胚胎成形素Nodal在喉癌组织中的表达情况及临床意义。方法 63例喉癌患者,取其喉癌组织(喉癌组)和癌旁正常组织(对照组),采用反转录-PCR检测Nodal mRNA表达水平,采用免疫组织化学检测Nodal蛋白表达情况和微血管密度,采用Western blot法检测Nodal蛋白表达水平,并分析Nodal表达水平与喉癌临床特征及微血管密度的关系。结果喉癌组Nodal mRNA(63.5±12.6)和蛋白(58.0±11.8)表达水平明显高于对照组(7.4±1.8、16.5±4.6)(P<0.01);喉癌患者Nodal mRNA和蛋白表达水平在TNM分期上差异有统计学意义(P<0.05),在性别、年龄上差异无统计学意义(P>0.05);随TNM分期增高,Nodal蛋白表达水平和微血管密度逐渐增强,Ⅳ期患者最高;不同临床分期喉癌组织中Nodal蛋白表达与相应微血管密度变化趋势呈正相关(r=0.485,P=0.035)。结论喉癌组织中Nodal mRNA和蛋白呈高表达,Nodal可能促进新生肿瘤血管形成而促进肿瘤发展。
Objective To explore the expression and significance of Nodal in laryngeal cancer.Methods Nodal mRNA level was detected by reverse transcription-PCR,the expression of Nodal protein and microvessel density were detected by immunohistochemistry,and Nodal protein level was detected by Western blot in laryngeal cancer tissue(cancer group)and normal adjacent tissue(control group)from 63 patients with laryngeal cancer.The relationship of Nodal expression with clinical characteristics and microvessel density changes was analyzed.Results The expressions of Nodal mRNA(63.5±12.6)and protein(58.0±11.8)in cancer group were significantly higher than those in control group(7.4±1.8,16.5±4.6)(P〈0.01).There were significant differences in the expression levels of Nodal mRNA and protein in TNM stages(P〈0.05),and no significant differences in the gender and age(P〈0.05).The Nodal protein expression level and microvessel density increased with TNM stages and were the highest in stageⅣ.The nodal protein expression was positively correlated with the tendency of microvessel density in different TNM stages(r=0.485,P=0.035).Conclusion Nodal mPNA and protein are highly expressed in laryngeal cancer tissue and could promote the cancer growth by facilitating the angiogenesis in neogenetic cancer.
