基质金属蛋白酶及其抑制物在月经周期中表达的分析 被引量:2

Analysis on the expressions of matrix metalloproteinases and tissue inhibitor of matrix metalloproteinases in menstrual cycle

摘要 目的观察子宫内膜组织中基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)和组织金属蛋白酶抑制物-1(TIMP-1)在正常月经周期中不同时期的表达,探讨其意义。方法月经正常者37例,子宫内膜组织分为增殖期、分泌早期、分泌中期,所有标本采用免疫组织化学法及RT-PCR技术检测MMP-2、MMP-9和TIMP-1的表达,并采用病理图像分析技术进行半定量分析。结果 MMP-9、MMP-2及TIMP-1在各组子宫内膜中均呈阳性表达,在腺上皮细胞和间质细胞胞浆均可见棕黄色颗粒沉着,以腺上皮细胞表达为著。分泌中期MMP-9、MMP-2及TIMP-1在子宫内膜腺上皮中的表达明显高于增殖期和分泌早期(P<0.05);分泌早期MMP-9、MMP-2及TIMP-1在子宫内膜腺上皮中的表达明显高于增殖期(P<0.05)。子宫内膜间质中的表达无明显差异(P>0.05)。结论人类月经周期中子宫内膜腺上皮中MMP-9、MMP-2及TIMP-1的表达呈周期性变化,其表达与子宫内膜功能相关。 Objective To observe the expressions of matrix metalloproteinase-2 (MMP-2), matrix metalloproteinase-9 (MMP-9), and tissue inhibitor of matrix metalloproteinases- 1 ( TIMP- 1 ) in normal menstrual cycle, explore the significance. Methods Thirty-seven women with normal menstruation were selected, their endometrial specimens were divided into proliferative phase, early secretory phase, and mid secretory phase. The expressions of MMP-2, MMP-9, and TIMP-1 in all the specimens were detected by immunohistochemistry and RT-PCR, pathological image analysis technology was used for semi-quantitative analysis. Results MMP-9, MMP-2, and TIMP-1 expressed positively in endometrial specimens in all the groups, brown granule pigmentation was found in glandular epithelial ceils and stromal cells, especially in the former ones. The expression levels of MMP-9, MMP-2, and TIMP-1 in glandular epithelial cells during mid secretory phase were statistically significantly higher than those during proliferative phase and early secretory phase ( P〈0.05 ) ; the expression levels of MMP-9, MMP-2, and TIMP-1 in glandular epithelial cells during early secretory phase were statistically significantly higher than those during proliferative phase (P〈0. 05 ) . There was no statistically significant difference in stromal cells of endometrium among the three groups (P〉0. 05) . Conclusion The cyclical expressions of MMP-9, MMP-2, and TIMP- 1 in human glandular epithelial cells of endometrium are correlated with endometrial function.
