摘要
目的:通过唾液酸糖基转移酶基因芯片检测ST3GalⅤ在人正常肝细胞系L-02及肝癌细胞系Hep G-2和SMMC-7721中的表达差异。方法:制备唾液酸糖基转移酶基因芯片,应用基因芯片检测ST3GalⅤ在人正常肝细胞系L-02及肝癌细胞系Hep G-2和SMMC-7721中的表达,并通过蛋白质印迹(Western blot)法检测蛋白表达水平。结果:唾液酸糖基转移酶ST3GalⅤ在人肝癌细胞系中表达降低。结论:ST3GalⅤ在肝癌细胞系中表达水平的降低导致神经节苷脂GD1a的合成下降从而影响肝细胞的生长增殖。
Objective: To investigate the differential expression of ST3Gal V in human hepatic carcinoma cell lines, HepG-2 and SMMC-7721 and normal hepatic cell line, L-02 by sialyltransferase DNA microarray. Methods: Expression of ST3Gal family was measured with DNA microarray. Western blot was used to verify DNA microarrays data. Result: ST3Gal V downregulated in HepG-2 and SMMC-7721 cell lines, compared with control cell line. Conclusion: The downregulation of ST3Gal V in HepG-2 and SMMC- 7721 cell lines may result in the reduction of GDla and influence the hepatocyte proliferations.
