模拟肾虚痰湿特征信息PCOS大鼠模型中GDF-9、BMPR-2的表达研究

摘要 目的:探讨生长分化因子-9(GDF-9)及骨形态发生蛋白受体-2(BMPR-2)在有肾虚痰湿特征信息的肥胖多囊卵巢综合征(PCOS)大鼠模型的卵巢颗粒细胞中的表达。方法:分别在来曲唑(Letrozole)和脱氢表雄酮(DHEA)造模基础上加用高脂乳剂,建立肥胖PCOS动物模型,结合阴道脱落细胞涂片、大鼠体质量增加幅度、血脂水平和卵巢形态学的改变进行模型验证,选用免疫组化法检测大鼠模型卵巢组织卵泡颗粒细胞中GDF-9、BMPR-2的表达。结果:模型建立成功,Letrozole模型组、Letrozole肥胖模型组GDP-9积分光密度总和均显著低于高脂模型组。此外,Letrozole肥胖模型组GDP-9平均灰度均值低于高脂模型组,DHEA模型组和DHEA肥胖模型组GDF-9积分光密度总和均显著高于空白对照组,Letrozole模型组BMPR-2积分光密度总和均显著高于高脂模型组,DHEA模型组BMPR-2积分光密度总和显著高于高脂模型组。结论:推测GDF-9、BMPR-2在肥胖PCOS大鼠模型卵巢的卵泡颗粒细胞中表达降低可能是肥胖PCOS发生发展的重要原因。 Objective: To study the expression of Growth differentiation factor 9( GDF-9) and Bone morphogenetic protein receptor type 2( BMPR-2) in the ovarian granular cells of the obese Polycystic Ovary Syndrome( PCOS) rat models which has the characteristics of the deficiency of kidney and the stagnation of phlegm-dampness. Method: The obesity PCOS rat models were built respectively with high fat emulsion on the basis of Letrozole and DHEA( Dehydroepiandrosterone) models. The models were verified by the smears of vaginal exfoliated cells,the weight growth rate when rats were killed,the lipid levels and the morphological changes of ovarian. Immunohistochemical method was chosen to detect the expression of GDF-2,BMPR-2 in the ovarian tissue granulosa cells of rat models. Results: Models were set up successfully. The GDP-9 integral optical density in Letrozole group and Letrozole obese group were both significantly lower than high fat emulsion group. Besides,the GDP-9 average grayscale valuein Letrozole obese group was lower than high fat emulsion group; the GDP-9 integral optical density in DHEA group and DHEA obese group were significantly higher than the blank control group; the BMPR-2 integral optical density in Letrozole group was significantly higher than high fat emulsion group; the BMPR-2integral optical density in DHEA group was significantly higher than high fat emulsion group,the differences were statistically significant. Conclusion: It can be inferred that the expression of GDF-9 and BMPR-2 reduced in the ovarian follicle granulosa cells of the obese PCOS rat models were likely to be the important reasons for occurrence and development of the obese PCOS.
