冠心病血瘀证的基因网络模块研究

摘要 目的:探讨冠心病血瘀证基因网络中重要模块的生物学功能与通路。方法:搜索文献中冠心病血瘀证相关基因,并运用CYTOSCAPE进行网络构建,MCODE进行模块划分后再对重要模块进行功能与通路分析。结果:冠心病血瘀证网络中BCL2、VEGFA、IL-6、JUN和IL-8具有重要作用。血管平滑肌增殖和toll样受体信号通路分别是网络中最重要的生物学过程与通路。结论:该研究既体现了冠心病血瘀证基因的整体特点,又关注了基因网络的局部差异,为冠心病的预防与治疗提供了新的思路。 Objective: Blood stasis is the predominant syndrome of coronary heart disease( CHD),however,there still lack of a genetic landscape of it. We are aiming to explore the biological processes and pathways of blood stasis of coronary heart disease. Methods: We obtained genes from the literatures and applied Agilent Literature Search for network construction,MCODE for modularized analysis. After that,a functional enrichment analysis was performed to find the biological processes and pathways of the most important modules. Results: 119 Blood stasis-related genes were found from the literatures and were used to mining more potential related genes. Totally,other 489 genes were found correlated to the blood stasis of CHD,612 nodes and 1834 edges consist of the whole network. Each node represents one gene,and edge means there is connection between the genes. Bcl2、VEGFA、IL-6、IL-8 and JUN play important roles in the network for they owning the most edges. Regulation of smooth muscle cell proliferation is the most important biological process of the significant module, toll-like receptor signaling is the most important pathway of it. Conclusion: Our findings are considerably in accord with previous research show that network analysis is an useful and stable tool. It allowed us to gain a better understanding of the pathological mechanism of the blood stasis of CHD.
