慢性HBV感染过程CD4^+T细胞TLR2表达水平的研究 被引量:2

摘要 目的:检测慢性乙型肝炎病毒(HBV)感染者CD4+T细胞中TLR2分子的表达与CD4+T细胞免疫活化与耗竭水平,研究HBV感染过程中CD4+T细胞TLR2表达的意义。方法:募集未经抗病毒治疗的HBV感染者40例,根据血清HBV病毒载量,分为高病毒载量组、低病毒载量组,另取19例健康体检者血液作为健康对照;使用流式细胞术检测HBV感染者CD4+T细胞亚群中TLR2与CD38、HLA-DR、CD95、PD-1分子的表达情况及其相关性。结果:低病毒载量组与高病毒载量组CD4+T细胞TLR2阳性率较健康对照组高,组间比较差异具有统计学意义(P<0.01);低病毒载量组CD4+T细胞亚群TLR2表达高于高病毒载量组,但组间比较差异无统计学差异(P=0.121 9);相关性分析结果显示CD4+T细胞亚群中TLR2与CD38、HLA-DR、CD95、PD-1表达无相关性(P>0.05)。结论:慢性HBV感染者CD4+T细胞TLR2的表达升高,提示TLR2在CD4+T细胞的表达可能参与了机体对HBV感染的免疫应答过程,但其具体机制还有待阐明。 Objective:In this study,we tested the TLR2 expression on CD4+T cells,and the activation and exhaustion levels of CD4+T cells,and to explore the mechanism of the TLR2 expression on CD4+T cells during HBV infection.Methods:40chronic HBV infected patients without anti-virus treatment enrolled in our study were divided into two groups including the high viral load group and the low viral load group,and19 healthy donors were recruited as the control group.After isolation of peripheral blood mononuclear cell(PBMC),we detected the expression of TLR2,CD38,HLA-DR,CD95 and PD-1on CD4+T cells by using flow cytometry.We also analyzed the correlation between TLR2 and CD38,HLA-DR,CD95,PD-1on CD4+T cells.Results:The expression of TLR2 on CD4+T cells in HBV infected patients are significantly higher than that in healthy donors(P 〈0.01).The TLR2 expression on CD4+T cells does not show significant difference between the patients with low viral load and patients with high viral load(P=0.121 9).Moreover,correlation analysis suggested that TLR2 expression is not associated with CD38,HLA-DR,CD95,and PD-1on CD4+T cells during chronic HBV infection.Conclusion:The increased expression of TLR2 on CD4+T cells in chronic HBV infected patients indicates that TLR2 may play an important role in HBV process,but the underlying mechanism still need further investigation.
