摘要
目的研究CpG 2006佐剂对呼吸道合胞病毒(RSV)重组蛋白疫苗诱导的体液免疫应答的作用。方法重组RSV疫苗G1F/M2与CpG佐剂混合,鼻腔或腹腔注射免疫BALB/c小鼠3次,最后一次免疫2周后取血,分离血清,用间接ELISA检测特异性抗体IgG、IgG1和IgG2a水平。结果鼻腔免疫途径:CpG佐剂组诱导的IgG1均显著低于无佐剂组(t=3.365,P<0.05),而IgG2a显著高于无佐剂组(t=2.675,P<0.05),佐剂组IgG1/IgG2a的比值(1.05)明显低于无佐剂组(1.25)。腹腔免疫途径:CpG佐剂组的IgG1明显低于无佐剂组(t=6.869,P<0.05),而IgG2a显著高于无佐剂组(t=2.893,P<0.05),且前者IgG1/IgG2a的比值(0.97)明显低于后者(1.30)。结论CpG 2006作为G1F/M2的佐剂,对体液免疫应答无增强作用,但可以调节免疫应答的类型,使Th1型应答升高,Th2型应答降低。
Objective To investigate the role of CpG 2006 adjuvant in the humoral immune responses induced by respiratory syncytial virus( RSV). Methods BALB / c mice were immunized i. n. or i. p,by intraperitoneal injection of nasal cavity or immune BALB / c mice 3 times,immunized mice were removed two weeks after the latest immunization. Specific antibodies IgG,IgG1 and IgG2 a were tested by indirect ELISA. Results The humoral immune response in mice immunized i. n. : IgG1 in CpG adjuvant groups was significantly lower than that in the group without adjuvant( t = 3. 365,P 〈0. 05),and IgG2 a was significantly higher than that in the group without adjuvant( t = 2. 675,P 〈0. 05). The ratio of IgG1 / IgG2 a of the CpG adjuvant groups( 1. 05) was obviously lower than the ratio of the group without adjuvant( 1. 25). The humoral immune response in mice immunized i. p. : The IgG1 was obviously lower than that of the group without adjuvant( t = 6. 869,P 〈0. 05),and IgG2 a was significantly higher than that in the group without adjuvant( t = 2. 893,P〈 0. 05). IgG1 / g G2 a ratio of the former( 0. 97) was obviously lower than that of the latter( 1. 30). Conclusion The CpG2006,as the single adjuvant of G1 F / M2,has no enhanced function on the humoral immune response,but it regulated the type of immune responses,increased the Th1 type response and decreased the Th2 type response.
